Key differences in regulation of opioid receptors localized to presynaptic terminals compared to somas: Relevance for novel therapeutics

Summary: Opioid receptors are G protein-coupled receptors (GPCRs) that regulate pain, reward, and aversion processing in the brain. They inhibit neurotransmitter release in presynaptic terminals and hyperpolarize neurons in postsynaptic locations. Activation of opioid receptors at the plasma membrane leads to regulatory processes such as phosphorylation and internalization of the receptors. The balance of opioid signaling in circuits expressing pre-and postsynaptic opioid receptors is shifted toward inhibition of presynaptic neurotransmitter release during continuous opioid exposure.

Summary: The search for new inhibitors of biologically relevant targets is an important strategy in developing new drugs for the treatment of neurodegenerative diseases. A series of compounds were designed, synthesized, and evaluated for their inhibitory potential against acetylcholinesterase and butyrylcholinesterase. One of the compounds showed the strongest inhibition activity against acetylcholinesterase and cytotoxicity studies were also conducted.

Antinociceptive Effects of VV-Hemorphin-5 Peptide Analogues Containing Amino phosphonate Moiety in Mouse Formalin Model of Pain

Summary: In this study, hemorphin analogues with amino phosphonate moiety were synthesized and tested for their antinociceptive activities in mice. The results showed that pVV-H and pLV-H analogues exhibited significant antinociceptive effects in acute and inflammatory pain, while pL-H and pLVV-H analogues were less effective. This study contributes to understanding the role of Valine and the number of amino acid residues in the effectiveness of hemorphin peptide analogs in suppressing experimental pain.
