Targeting the STAT5A/IDO1 axis overcomes radioresistance and reverses the immunosuppressive tumor microenvironment in NSCLC

Yang Yang et al.

Summary: This study investigated the role of IDO1 in the NSCLC microenvironment and found that targeting the STAT5A/IDO1 axis can reshape the immune microenvironment and promote the efficacy of RT in NSCLC treatment.

An adverse tumor-protective effect of IDO1 inhibition

Juliana C. N. Kenski et al.

Summary: By restoring tryptophan, IDO1 inhibitors aim to reactivate anti-tumor T cells. However, inhibiting IDO1 can lead to an adverse protection of melanoma cells to T cell-derived IFNg. This is accompanied by impaired translation and a stress response that downregulates MITF, contributing to T cell resistance and potentially impacting patient outcome.

Cancer-associated adipocytes promote the invasion and metastasis in breast cancer through LIF/CXCLs positive feedback loop

Chong Zhou et al.

Summary: Cancer-associated adipocytes (CAAs) and breast cancer cells interact with each other through the secretion of cytokines. CAAs secrete leukemia inhibitory factor (LIF), which promotes migration and invasion of breast cancer cells through the Stat3 signaling pathway, while breast cancer cells secrete C-X-C subfamily chemokines (CXCLs) to induce the expression of LIF in CAAs. This positive feedback loop between LIF and CXCLs promotes breast cancer invasion and metastasis.

Combination of IDO1(high) and CCL19(low) expression in the tumor tissue reduces survival in HPV positive cervical cancer

Jayakumar Hascitha et al.

Summary: In cervical cancer, overexpression of IDO1 and elevated K/T levels were found in early and locally advanced stages. High CCL19 marker expression was identified as a good prognostic indicator for disease-free and overall survival. High levels of IDO1 expression may further reduce survival outcomes in patients with low CCL19 expression.

Combined Inhibition of SHP2 and CXCR1/2 Promotes Antitumor T-cell Response in NSCLC

Kwan Ho Tang et al.

Summary: Our study suggests that inhibiting the SHP2/RAS/ERK pathway leads to upregulation of CXCR2 ligands and recruitment of S100A8 hi gMDSCs, suppressing T cells in non-small cell lung cancer. Combining SHP2/CXCR2 inhibitors can block gMDSC immigration, enhance Th1 polarization, induce highly cytotoxic CD8 + KLRG1 + effector T cells, and improve survival.

Jiping Sun et al.

Summary: This study identified CXCL1 as a key prognostic gene for cervical cancer by analyzing gene expression data and clinicopathological data. The CXCL1/CXCR2 chemokine system was found to regulate the proliferation, migration, and apoptosis of cervical cancer cells through an autocrine loop, thereby affecting the development of cervical cancer.

Roberta Liberato Pagni et al.

Summary: The study demonstrated that IL-6 and IDO negatively contribute to the activation of immune cells, particularly dendritic cells, reducing protective immune responses induced by the gDE7 vaccine. Therefore, combined strategies based on inhibition of IL-6 and IDO offer new perspectives for immunotherapy against HPV-related tumors.

HOXC10 Promotes Metastasis in Colorectal Cancer by Recruiting Myeloid-derived Suppressor Cells

Jiao Yu et al.

Summary: This study found that HOXC10 is associated with metastasis in CRC patients and has prognostic value. HOXC10 overexpression enhances the metastatic ability of CRC cells and promotes the infiltration of MDSCs by upregulating CXCL5. CXCR2 inhibitors and anti-PD-L1 can effectively prevent CRC metastasis.

Summary: Tumor recurrence in GBM is a major challenge, with upregulation of IL8/CXCL2/CXCR2 signaling induced by TMZ. High TAM infiltration predicts poor survival, while more patients express IL8 in recurrent tumors and TMZ therapy maintains CXCL2 expression. Combination therapy shows enhanced anti-tumoral effects.
