Laponite-Based Nanocomposite Hydrogels for Drug Delivery Applications

Aptamer-immobilized bone-targeting nanoparticles in situ reduce sclerostin for osteoporosis treatment

Summary: In this study, a novel bone-targeted nanomedicine was developed for the treatment of osteoporosis. The nanomedicine demonstrated dual functions of directing bone attachment and capturing sclerostin with high affinity. The results showed that the nanomedicine significantly reversed serum levels of sclerostin and bone loss associated with osteoporosis, improving bone histomorphology parameters and mechanical properties in the femur. Furthermore, the therapeutic effect of the nanomedicine was superior to the gold standard drug alendronate, while requiring only 25% of the systemic dose.

Bone regeneration in rat calvarial defects using dissociated or spheroid mesenchymal stromal cells in scaffold-hydrogel constructs

Summary: Three-dimensional spheroid culture can promote osteogenic differentiation of bone marrow mesenchymal stromal cells, while 3D printing allows for customized scaffolds for complex bone defects. Results showed a trend for superior in vitro mineralization with 3D BMSC, but similar in vivo performance between 2D and 3D BMSC, indicating PLATMC-HPLG constructs as promising scaffolds for bone tissue engineering.
