Xuewei Zhang et al.

Summary: The application of decellularized extracellular matrix (dECM) scaffolds holds enormous potential for tissue regeneration. These scaffolds provide an optimal non-immune environment with native three-dimensional structures and bioactive components, making them an ideal choice for regenerating functional organs/tissues, especially when combined with stem cells.

Reprogramming of fibroblasts into expandable cardiovascular progenitor cells via small molecules in xeno-free conditions

Jia Wang et al.

Summary: A combination of small molecules can generate a cell population with cardiovascular differentiation potential from fibroblasts. These chemically induced cardiovascular progenitor cells (ciCPCs) can self-renew and preserve their differentiation capacity in chemically defined and xeno-free conditions. Transplantation of ciCPCs into infarcted mouse hearts improves cardiac function. Expanded autologous cardiovascular cells have important applications in drug discovery, disease modeling, and cardiac cell therapy.

Case report: Transplantation of human induced pluripotent stem cell-derived cardiomyocyte patches for ischemic cardiomyopathy

Shigeru Miyagawa et al.

Summary: Despite being a life-threatening disease, heart failure can potentially be treated using allogenic human induced pluripotent stem cell-derived cardiomyocyte (hiPSC-CM) patches. The successful implantation of these patches in a patient with severe ischemic cardiomyopathy showed potential efficacy, safety, and tolerability, improving clinical symptoms and recovering wall motion at the transplanted site.

Cardiomyocytes induced from hiPSCs by well-defined compounds have therapeutic potential in heart failure by secreting PDGF-BB

Hongmei Li et al.

Summary: Recent studies have shown that transplantation of hiPS-CMs is a promising approach for treating heart failure, but the immature nature and secreted proteins of hiPS-CMs involved in cardiomyocyte repair are not well understood. In this study, a saponin(+) compound induced system was established to generate stable and functional hiPS-CMs in vitro and transplanted into heart failure mice. The results demonstrated that the optimally induced hiPS-CMs had enhanced therapeutic effects by reducing cardiac remodeling and dysfunction, decreasing cardiomyocyte death, and promoting angiogenesis. The reparative effect was likely mediated by the secretion of PDGF-BB and the activation of the PI3K/Akt pathway.

Angiogenesis after acute myocardial infarction

Xuekun Wu et al.

Summary: The translation describes how acute myocardial infarction causes damage to the coronary microcirculation, leading to vascular disintegration and capillary rarefication in the infarct region. Tissue repair after MI involves robust angiogenic response and interactions among various cell types through secreted proteins and receptors. Macrophages and fibroblasts have emerged as major drivers of the angiogenic response after MI, while understanding the mechanism of infarct angiogenesis creates therapeutic opportunities.

Strontium ions protect hearts against myocardial ischemia/reperfusion injury

Min Xing et al.

Summary: The injection of composite hydrogels containing strontium ions into infarcted myocardium after ischemia and reperfusion in mice significantly attenuated functional deterioration and scar formation, while also reducing cardiomyocyte apoptosis and promoting angiogenesis. These findings suggest the cardioprotective effects of strontium ions against ischemia/reperfusion injury, providing a potential new therapeutic approach for ischemic heart disease.

Neurotrophin-3 contributes to benefits of human embryonic stem cell-derived cardiovascular progenitor cells against reperfused myocardial infarction

Wei Bi et al.

Summary: This study demonstrates that NT-3 secreted by hCVPCs plays a cardioprotective role by reducing cardiomyocyte apoptosis and promoting angiogenesis. Furthermore, NT-3 inhibits I/R-induced cardiomyocyte apoptosis via the ERK-Bim signaling pathway. NT-3 may serve as a noncell approach for the treatment of myocardial I/R injury.

The Present State and Future Perspectives of Cardiac Regenerative Therapy Using Human Pluripotent Stem Cells

Yusuke Soma et al.

Summary: The number of patients with heart failure is increasing due to global aging, and heart transplantation is a treatment option for drug-resistant HF patients, but donor heart shortages are a serious issue. Cardiac regenerative therapy using hPSCs is expected to be an alternative to HT, but post-transplantation issues such as arrhythmia and immune rejection need to be addressed.

Myosin light chain 2 marks differentiating ventricular cardiomyocytes derived from human embryonic stem cells

Xiao-Ling Luo et al.

Summary: Human pluripotent stem cell-derived cardiomyocytes (hPSC-CMs) have great value in various studies, but their heterogeneity and maturation inconsistency have limited their applications. Myosin light chain 2 (MYL2) has been identified as a marker for ventricular cardiomyocytes, but its role in hESC-CMs has been questioned. This study revealed a correlation between MYL2 expression and the development of ventricular hESC-CMs, shedding light on the development of human ventricular myocytes and providing insights into overcoming hESC-CM heterogeneity.
