Sensor Selection and State Estimation of Continuous mAb Production Processes

Process-aware data-driven modelling and model predictive control of bioreactor for the production of monoclonal antibodies

Summary: This manuscript addresses the issue of controlling a bioreactor to optimize the production of a desired product while complying with the constraints imposed by the bio-process. It proposes a data-driven model and a model predictive controller (MPC) to achieve this goal, which are demonstrated using a monoclonal antibody production model. The study incorporates known physical relationships in the data-driven model development through constrained subspace model identification, resulting in improved production efficiency compared to the existing proportional integral (PI) controller strategy. Simulation results also show the superiority of the process-aware or constrained subspace MPC over traditional subspace MPC. Furthermore, the robustness of the controller design is illustrated by its ability to maintain good performance even with changes in cell line or feed characteristics.

A sensitivity-based approach to optimal sensor selection for complex processes

Summary: This paper presents a sensitivity-based approach for determining the minimum number of sensors and their optimal locations for state estimation. The minimum number of sensors required to achieve full column rank is determined based on the local sensitivity matrix of the output measurements to initial states. The optimal sensor placement is the sensor set that provides the maximum degree of observability among all sets that meet the full-rank requirement. The computational complexity of sensor selection is significantly reduced by successive orthogonalization of the columns of the sensitivity matrix. The effectiveness of the proposed method is validated through application to two processes: four continuous stirred-tank reactors and a wastewater treatment plant. In both cases, the proposed approach can obtain the optimal sensor subset.
