Summary: The in vitro production of functional gametes, particularly sperm, from premeiotic spermatogonia harvested from marine four-eyed sleepers has been successful in this study. Melatonin was found to significantly promote spermatogonia proliferation and differentiation through the ERK1/2 signaling pathway, leading to increased efficiency in functional sperm production. This 3D culture system shows great promise for enhancing genetic breeding in aquaculture fish.

Summary: In vitro spermatogenesis for marine medaka can be performed via whole testis organ cultures, but maintaining spermatogenesis during the early phase of culturing requires further optimization of culture conditions. Modifying factors such as culture temperature, basal media, and medium supplements can significantly impact spermatogonial proliferation. The optimized culture conditions using alpha-MEM and KSR were found to have the most positive influence on maintaining spermatogonial proliferation levels in whole testis organ cultures.
