Erythrocytosis, thrombocytosis, and rate of recurrent thromboembolic event—A population based cohort study

Summary: Management strategies for reducing the risk of thromboembolic complications in polycythemia vera and essential thrombocythemia are well established, but for other conditions with elevated blood values, there is no consensus on treatment and follow-up. This retrospective cohort study of 3931 adult patients in Sweden found that elevated blood values are common in patients with thromboembolic events and the risk of recurrence and restricted mean survival time is highest in patients with unexplained thrombocytosis and secondary erythrocytosis. This highlights the importance of identifying and managing the underlying condition.

Downregulation of miR-125a-5p Promotes Endothelial Progenitor Cell Migration and Angiogenesis and Alleviates Deep Vein Thrombosis in Mice Via Upregulation of MCL-1

Summary: This study aimed to investigate the effect of miR-125a-5p on endothelial progenitor cells (EPCs) in deep vein thrombosis (DVT). It was found that upregulation of miR-125a-5p suppressed EPC migration and angiogenesis, while promoting apoptosis. Downregulation of miR-125a-5p had the opposite effect. Furthermore, miR-125a-5p inhibited the PI3K/AKT pathway. Cellular experiments showed that MCL-1 was regulated by miR-125a-5p. Animal experiments demonstrated that downregulation of miR-125a-5p inhibited thrombus formation and enhanced EPC homing, thereby alleviating DVT.

Exosomal miR-145 and miR-885 Regulate Thrombosis in COVID-19

Summary: We hypothesized that exosomal microRNAs may play a role in the development of thromboembolic complications in COVID-19. We isolated circulating exosomes from COVID-19 patients and categorized them based on D-dimer levels. We found that exosomal miR-145 and miR-885 were significantly correlated with D-dimer levels. Furthermore, human endothelial cells treated with serum from COVID-19 patients showed reduced release of miR-145 and miR-885, increased apoptosis, and impaired angiogenetic properties compared to cells treated with non-COVID-19 serum.

In this study, we found that circulating small extracellular vesicles (sEVs) were increased in PAPS patients, mainly originating from activated platelets. These sEVs contained specific miRNAs, including miR-483-3p and miR-326, which were associated with endothelial cell proliferation and monocyte activation, respectively. Our findings suggest that miRNA-derived sEVs play a role in the pathogenesis of APS.

Summary: This study found that levels of hypoxia-inducible factor-1 alpha (HIF-1 alpha) were significantly higher in patients with transplantation-associated thrombotic microangiopathy (TA-TMA) compared to controls. Upregulation of HIF-1 alpha led to an increase in membrane-bound complement C3 and dysfunction of endothelial cells. Increasing HIF-1 alpha in mice led to complement activation, thrombocytopenia, anemia, and increased lactate dehydrogenase (LDH) levels, mimicking the TA-TMA phenotype. The study suggests that HIF-1 alpha may play a role in the development of TA-TMA by inhibiting the transcription of complement factor H (CFH).

The combination of miR-448 and SIRT1 has a high predictive value for the occurrence of DVT.
