Development of a Simple DNA Extraction Method and Candida Pan Loop-Mediated Isothermal Amplification Assay for Diagnosis of Candidemia

Summary: To reduce the morbidity and mortality of candidemia patients, a simple and rapid molecular diagnostic method based on nucleic acid extraction is needed. We developed a multiplex Candida Pan/internal control (IC) loop-mediated isothermal amplification (LAMP) assay and a Chelex-100 boiling protocol for DNA extraction. The results showed high sensitivity and specificity, suggesting that this method could be used as a rapid molecular diagnostic test for candidemia.

This study combines heat deactivation and mechanical lysis by bead beating for simple and quick sample preparation, followed by an optimized RT-LAMP assay targeting specific genes of the novel coronavirus. The system shows high diagnostic performance and faster time-to-result compared to commercial RT-PCR kits.
