Leveraging molecular structure and bioactivity with chemical language models for de novo drug design

Michael Moret et al.

Summary: Generative chemical language models (CLMs) can be used to generate new molecular structures from a textual representation. Hybrid CLMs can leverage bioactivity information for training compounds. In this study, a virtual compound library was created using a generative CLM and refined using a CLM-based classifier for bioactivity prediction. A new PI3K gamma ligand with sub-micromolar activity was identified, highlighting the potential of hybrid CLMs for molecular design.

ZINC-22-A Free Multi-Billion-Scale Database of Tangible Compounds for Ligand Discovery

Benjamin I. Tingle et al.

Summary: The purchasable chemical space has expanded rapidly to billions of molecules, presenting unprecedented opportunities for ligand discovery but also posing challenges for scalable tools. To address this, ZINC-22, a database of commercially available small molecules derived from large-scale make-on-demand libraries, has been developed. The new database and tools enable analog searching in this vast chemical space efficiently and provide access to physical properties crucial for molecule docking. The diversity of molecules continues to grow with the database size, and the implications of the increasing library size are considered.

Artificial intelligence-enabled virtual screening of ultra-large chemical libraries with deep docking

Francesco Gentile et al.

Summary: With the rapid expansion of chemical libraries, more efficient virtual screening approaches are needed. The Deep Docking (DD) platform allows for accelerated structure-based virtual screening by docking only a subset of the library and synchronizing it with ligand-based predictions. This method enables the screening of large chemical libraries without requiring extraordinary computational resources.

A Consensus Compound/Bioactivity Dataset for Data-Driven Drug Design and Chemogenomics

Laura Isigkeit et al.

Summary: Publicly available compound and bioactivity databases are essential for data-driven applications in life-science research and drug design. This study analyzed multiple bioactivity repositories and found differences in compound and target coverage, highlighting the importance of using data from multiple sources. By combining data from various databases, a consensus dataset with improved compound and target coverage was assembled. This dataset included over 1.1 million compounds and over 10.9 million bioactivity data points, providing a valuable resource for computational applications in drug design and chemogenomics.

Synthesis, Characterisation and Mechanism of Action of Anticancer 3-Fluoroazetidin-2-ones

Azizah M. Malebari et al.

Summary: This study describes the synthesis, characterization, and mechanism of action of a series of 3-fluoro and 3,3-difluoro substituted beta-lactams as analogues of the tubulin-targeting agent CA-4. These compounds showed potent activity in breast cancer cells and low toxicity in non-cancerous cells. Mechanistic studies demonstrated that these compounds inhibit tubulin polymerization and induce apoptosis, making them potential candidates for the development of novel cancer therapeutics.
