Higher-order cycloadditions in the age of catalysis

Nicolaj Inunnguaq Jessen et al.

Summary: This article discusses the classification and analysis of successful organocatalytic higher-order cycloadditions (HOCs). Aminocatalysis is the most commonly used activation strategy, and the lessons learned from it can contribute to the development of other organocatalytic strategies. The potential for continued growth of catalytic HOCs as a research field is demonstrated by sporadic examples.

Enantioselective (8+3) Cycloadditions by Activation of Donor-Acceptor Cyclopropanes Employing Chiral Bronsted Base Catalysis

David A. McLeod et al.

Summary: This study presents a novel enantioselective (8+3) cycloaddition reaction using a chiral bifunctional Bronsted base catalyst. The reaction leverages an anionic activation strategy and differs from typical Lewis-acid activation protocols. Experimental and computational results support the relevance of this reaction and its potential application in organic synthesis.

Unveiling the Chemistry of Higher-Order Cycloaddition Reactions within the Molecular Electron Density Theory

Luis R. Domingo et al.

Summary: The higher-order cycloaddition reaction between tropone and cyclopentadiene has been studied, revealing the electronic structure and reaction pathways. The reaction exhibits high stereoselectivity, pseudocyclic selectivity, and regioselectivity due to favorable nucleophilic attacks and electrostatic interactions.

Understanding the higher-order cycloaddition reactions of heptafulvene, tropone, and its nitrogen derivatives, with electrophilic and nucleophilic ethylenes inside the molecular electron density theory

Luis R. Domingo et al.

Summary: This study investigated the higher-order cycloaddition reactions of four cycloheptatriene (CHT) derivatives using molecular electron density theory (MEDT). The results showed that the substitution at the X8 position of these CHTs only affected the electron density of the C1-X8 bonding region, and the CHT core was only slightly polarized. Heptafulvene and the two nitrogen derivatives were classified as strong nucleophiles, while tropone and the ylidenehydrazine were classified as strong electrophiles. The reactions with dicyanoethylene (DCE) were completely regioselective, while the reactions with cyclic ketene acetal (CKA) were highly regio- and pseudocyclic selective for heptafulvene and the imine, and tropone and ylidenehydrazine, respectively. MDET analysis provided insights into the reactivity and selectivity of these higher-order cycloaddition reactions based on the substitution at the X8 center of these CHTs.

[8+2] vs [4+2] Cycloadditions of Cyclohexadienamines to Tropone and Heptafulvenes-Mechanisms and Selectivities

Xiangyang Chen et al.

Summary: The study investigates the selectivity of [8+2] and [4+2] cycloadducts in cinchona alkaloid-catalyzed cycloaddition reactions, examining the factors influencing these pathways. It was found that the formation of final products is influenced by the reversibility of the reaction and the thermal stability of the products.

A molecular electron density theory study of the higher-order cycloaddition reactions of tropone with electron-rich ethylenes. The role of the Lewis acid catalyst in the mechanism and pseudocyclic selectivity

Summary: The study demonstrates that the higher-order cycloaddition reactions of tropone with nucleophilic ethylenes, catalyzed by Lewis acids, proceed through a non-concerted two-stage one-step or a two-step mechanism, resulting in the formation of a single cycloadduct.

Three-Component, Diastereoselective [6+3] Annulation of Tropone, Imino Esters, and Arynes

Avishek Guin et al.

Summary: This study demonstrates a transition-metal-free, three-component, and diastereoselective [6 + 3] annulation reaction using tropone, imino esters, and arynes to synthesize bridged azabicyclo[4.3.1]decadienes. The nitrogen ylides generated from imino esters and arynes undergo regioselective addition to tropone to produce the desired products with moderate to good yields and good functional group tolerance under mild conditions.
