Intracellular Reactive Oxygen Species-Aided Localized Cell Death Contributing to Immune Responses Against Wheat Powdery Mildew Pathogen

Summary: Reactive oxygen species (ROS) and hypersensitive response (HR) play important roles in plant immunity against pathogens. In this study, we quantitatively analyzed the accumulation of apoplastic ROS (apoROS) and intracellular ROS (intraROS) in wheat cells infected with Blumeria graminis f. sp. tritici (Bgt). We found that apoROS accumulation accounted for 70 to 80% of infected wheat cells in both compatible and incompatible host-pathogen interactions. IntraROS accumulation and localized cell death responses were mainly observed in wheat lines carrying nucleotide-binding leucine-rich repeat R genes. Our results provide new insights into the immune responses against wheat powdery mildew.

Molecular characterization and validation of adult-plant stripe rust resistance gene Yr86 in Chinese wheat cultivar Zhongmai 895

Summary: The adult-plant stripe rust resistance gene, Yr86, in Chinese wheat cultivar Zhongmai 895 was mapped to the physical interval 710.2-713.2 Mb on chromosome 2A. This gene provides stable resistance to stripe rust at the adult-plant stage and has the potential to be a new gene for this type of resistance. Multiple KASP markers associated with Yr86 were developed and can be used for marker-assisted selection and further studies on this resistance gene.

Stripe rust resistance gene(s) postulation in wheat germplasm with the help of differentials and tagged molecular markers

Summary: Thirteen known Yr gene-associated markers were used to identify the resistance genes in selected wheat germplasm against stripe rust. High resistance was observed in 38 genotypes, while seven genotypes showed resistant to moderately resistant response. In race-specific phenotyping, 29.2% genotypes were immune, 58.3% were resistant, and 6.3% were moderately resistant. Yr5, Yr10, Yr15, and Yr24/26 were detected in different genotypes using specific markers. The frequencies of Yr5, Yr15, and Yr24/26 were higher compared to Yr10 in the tested wheat germplasm.

Multi-environment phenotyping to identify broad-based, stable resistance in wheat germplasms against leaf and stripe rust diseases

Summary: Globally, leaf and yellow rusts are major biotic constraints in wheat production in India, causing significant economic losses. Host resistance is the most effective and eco-friendly approach to control rust diseases. This study characterized the resistance of different wheat genotypes against rust diseases, identifying multiple Yr and Lr genes in Indian wheat germplasm accessions and cultivars. Promising adult plant slow rusting resistance was observed, and epidemiological parameters were correlated, highlighting the appropriate parameters to assess resistance. The identified rust-resistant genotypes are valuable for enhancing resistance levels in hybridization programs.

Pre-anthesis spike growth dynamics and its association to yield components among elite bread wheat cultivars (Triticum aestivum L. spp.) under Mediterranean climate

Summary: This study examined bread wheat cultivars under Mediterranean semi-arid conditions and found significant variability in the timing of pre-anthesis phases. The duration of leaf and spikelet initiation, as well as stem elongation, had an impact on spike-related traits and biomass partitioning. The duration of these pre-anthesis phases plays a crucial role in wheat yield enhancement.

Screening of different wheat genotypes against leaf rust and role of environmental factors affecting disease development

Summary: Leaf rust caused by Puccinia has a higher potential to threaten wheat yield compared to stem and yellow rust, with a potential loss of up to 10%. The severity of the disease depends on plant development and susceptibility. Breeding resistant varieties is the best and economic way to control this problem. Only a few varieties have shown resistance against leaf rust in the past three years. Low temperature, high humidity, and heavy rainfall promote disease development.

Marker-Assisted Improvement of Bread Wheat Variety HD2967 for Leaf and Stripe Rust Resistance

Summary: The mega wheat variety HD2967 was improved for leaf and stripe rust resistance through marker-assisted backcross breeding. The introduction of the leaf rust resistance gene LrTrk from the durum wheat genotype Trinakria and the use of linked and polymorphic markers facilitated the selection process. The resulting NILs showed resistance to both leaf and stripe rust, with comparable yields to the recurrent parent variety HD2967.

Capturing Wheat Phenotypes at the Genome Level

Summary: Recent technological advances in next-generation sequencing have made DNA sequencing more accessible for species with large and complex genomes, including bread wheat. Despite its importance as a food crop, breeding in bread wheat has been limited by its large polyploid genome. However, after years of effort, draft and reference genome sequences of bread wheat have been released, providing valuable resources for breeding. These advances have also facilitated the use of genotyping arrays in wheat breeding, allowing for fast and reliable characterization of wheat lines. Additionally, gene-editing and genomic selection methods have shown promise in improving important traits in wheat breeding.

Transfer of Durable Stripe Rust Resistance Gene Yr39 into Four Chinese Elite Wheat Cultivars Using Marker-Assisted Selection

Summary: In this study, the wheat gene Yr39 was successfully transferred into four Chinese elite wheat cultivars, resulting in the development of 11 selected lines with stripe rust resistance and desirable agronomic traits. Identification of Stripe Rust Resistance Genes in Common Wheat Cultivars and Breeding Lines from Kazakhstan
