Long non-coding RNAs: definitions, functions, challenges and recommendations

LINC01711 promotes transforming growth factor-beta (TGF-β) induced invasion in glioblastoma multiforme (GBM) by acting as a competing endogenous RNA for miR-34a and promoting ZEB1 expression

Summary: GBM is an aggressive and malignant tumor in the central nervous system. TGF-beta expression is elevated in GBM and promotes invasion and EMT. Several lncRNAs regulated by TGF-beta promote glioma pathogenesis, including LINC01711. LINC01711 is significantly upregulated in GBM tissues and associated with poor overall survival in patients, and it promotes proliferation, migration, and invasion in GBM. Depletion of LINC01711 sensitizes glioma cells to TMZ-induced apoptosis by inhibiting ZEB1 expression, and it functions as a competing endogenous RNA for miR-34a to promote ZEB1 expression and regulate invasion. LINC01711 appears to be a promising therapeutic target for GBM.

Silencing specific transcription factors suppressed tumor growth, suggesting a potential therapeutic target for addressing the challenges associated with intratumoral heterogeneity.

Circular RNAs: implications of signaling pathways and bioinformatics in human cancer

Summary: Circular RNAs (circRNAs), widely expressed in eukaryotic cells, play important roles in post-transcriptional control of gene expression and have multiple functions in biological processes. They serve as microRNA sponges, RNA-binding proteins, and templates for translation. CircRNAs are also involved in cancer progression and show potential as biomarkers for tumor diagnosis and therapy. Computational models, signaling pathway data, and bioinformatics databases have been used to explore circRNA-disease associations. This review focuses on the biological characteristics and functions of circRNAs, with a specific emphasis on their roles in cancer and the status of circRNA-associated bioinformatics databases. The potential of circRNAs as prognostic biomarkers in cancer is also discussed.

The role of UCA1 and WRAP53 in diagnosis of hepatocellular carcinoma: A single-center case-control study

Summary: The aim of this study was to investigate the diagnostic potential of serum urothelial carcinoma-associated 1 (UCA1) and wd repeat containing antisense to TP53 (WRAP53) as biomarkers for hepatocellular carcinoma (HCC). The results showed that both UCA1 and WRAP53 were upregulated in HCC patients compared to patients with liver cirrhosis and healthy controls. The combination of UCA1, WRAP53, and AFP improved the diagnostic performance for HCC. In conclusion, serum UCA1 and WRAP53 have the potential to be used as non-invasive diagnostic biomarkers for HCC with high sensitivity and specificity.

Profiling and Bioinformatics Analyses of Differential Circular RNA Expression in Glioblastoma Multiforme Cells Under Hypoxia

It identified 672 dysregulated circRNAs in LN229 and 698 dysregulated circRNAs in T98G. These differentially expressed circRNAs were found to be associated with GBM cell biogenesis and metabolism, as well as pathways such as TGF-beta signaling and HIF-1 signaling. Inhibition of hsa_circ_0000745 had a significant impact on GBM cell proliferation, migration, and invasion. Additionally, the study revealed that these circRNAs could interact with related miRNAs. These findings suggest that circRNAs play a role in GBM development and may have potential as prognostic markers and adjuvant therapeutics.
