Advances of Antimicrobial Peptide-Based Biomaterials for the Treatment of Bacterial Infections

Summary: Due to the increase in multidrug-resistant bacterial isolates and lack of effective antimicrobial agents, antibiotic resistant bacterial infections have risen significantly. There is an urgent need to develop new antimicrobial drugs and formulations. Recent advancements in natural antimicrobial peptides (AMPs), such as AMP optimization, self-assembled AMPs, AMP hydrogels, and biomaterial-assisted delivery, have shown promise in treating bacterial infections. This review focuses on the development prospects and limitations of AMP-based biomaterials for animal model infections like abdominal, skin, and eye infections, aiming to inspire further innovation and accelerate their commercialization.

Characteristics of Hybrid Bioglass-Chitosan Coatings on the Plasma Activated PEEK Polymer

Summary: Polyetheretherketone (PEEK) is a biocompatible polymer that has similar properties to human bone, making it a desirable material for orthopedic implants. However, PEEK lacks bioactivity due to its biological inertness. Surface modification and composite preparation are two strategies used to improve the bioactivity of PEEK. In this study, plasma activation and solution dip-coating techniques were used to embed bioglass and chitosan onto PEEK surfaces, resulting in changes in surface properties and improved bioactivity.

A multi-technique and multi-scale analysis of the thermal degradation of PEEK in laser heating

Summary: The present work investigates the thermal degradation of laser-heated PEEK and its effects on morphology, composition, crystallinity, and mechanical properties. Various methods including microscopy, spectroscopy, and nanoindentation are employed to examine the thermal damage. The results demonstrate that short-time laser heating can improve the crystallinity and hardness of PEEK, while prolonged heating leads to surface carbonisation and the formation of a char layer. The char layer acts as a protective mechanism, resulting in mainly superficial thermally induced damage.

Design and Synthesis of Novel Antimicrobial Agents

Summary: The need for innovative antimicrobials has increased due to the spread of multidrug-resistant bacteria. Overuse, abuse, and misuse of antibiotics are linked to antibiotic resistance. This paper reviews various approaches to develop novel classes of antibiotics with no pre-existing resistance in bacterial pathogens. These include nanotechnology, computational methods, antibiotic alternatives, drug repurposing, synthesis of novel antibacterial agents, and prodrugs. Regulations, public education, research, and technological advancements are crucial in combating antibiotic resistance.

A polypeptide coating for preventing biofilm on implants by inhibiting antibiotic resistance genes

Summary: Aging population has increased the demand for orthopedic implants. However, biofilm formation on implants due to antibiotic insensitivity and antimicrobial resistance has been a major challenge. In this study, an antibacterial polypeptide coating was developed to prevent biofilm formation and acquired antibiotic resistance. The coating demonstrated excellent in vivo adhesive capacity and strong antibacterial activity. It also inhibited the expression of genes associated with antimicrobial resistance. In vivo experiments confirmed the effectiveness of the coating in preventing biofilm formation and infections. Overall, this study provides a simple method for preparing antibacterial orthopedic implants without inducing antimicrobial resistance.

Blending strategy to modify PEEK-based orthopedic implants

Summary: Polyetheretherketone (PEEK) is a thermoplastic polymer widely used in orthopedic implants due to its strong mechanical properties, good chemical stability, and excellent biocompatibility. However, its low elastic modulus limits its application to low-modulus areas of human bone. This review discusses various strategies to modify PEEK-based implants, including blending with reinforcing materials to enhance mechanical strength and incorporating bioactive additives to promote osteogenic activity and bone integration. The challenges and future development of PEEK-based composites are also highlighted.

This review discusses the severity of diseases caused by microbial biofilm formation and explores the use of AMPs and their mechanisms against microbial biofilms, along with strategies to enhance their activity.

Summary: PEEK is a promising material in the fields of oral repair and orthopedic implantation due to its stability and proper elastic modulus. In this study, a sulfonated 3D porous PEEK decorated with Ag+ ions loaded ZIF-8 showed effective antibacterial performance against E. coli and S. aureus. The synergistic effect of Ag+ and Zn2+ ions released continuously from SPZA contributes to its excellent antimicrobial activity, providing a potential candidate for biomedical implants.

Enhancing antibacterial capability and osseointegration of polyetheretherketone (PEEK) implants by dual-functional surface modification

Summary: The study successfully developed a dual-functional PEEK implant surface that can simultaneously resist bacterial contamination and promote osseointegration by releasing MOX and OGP. The surface exhibited excellent antibacterial effects against Staphylococcus aureus and Escherichia coli, and significantly enhanced cell adhesion, proliferation, and osteogenicity. Animal models confirmed the in vivo antimicrobial and osteogenesis abilities of the dual-functional PEEK surface.
