Nanomedicines and nanocarriers in clinical trials: surfing through regulatory requirements and physico-chemical critical quality attributes

Diego Alejandro Dri et al.

Summary: Elucidating the physical-chemical characteristics of investigational medicinal products requires appropriate methodology. Due to the complexity and innovative nature of nanomedicines and nanocarriers, additional specific properties need to be defined for their characterization in clinical trials. The availability of regulatory requirements and guidelines for critical quality attributes of these nanotechnologies in a clinical trial setting is investigated, while also addressing current challenges and issues. Efforts from a regulatory standpoint are needed to ensure standardized characterization, streamline the approval process, and support the development of nanomedicines and nanocarriers in clinical drug trials.

Quantifying Chemical Composition and Reaction Kinetics of Individual Colloidally Dispersed Nanoparticles

Nathan D. Donahue et al.

Summary: By using dual analyte single-particle inductively coupled plasma quadrupole mass spectrometry, researchers were able to quantify the mass and chemical composition changes of individual colloidal nanoparticles during two different chemical reactions. This study revealed the heterogeneity of chemical reactions at the single nanoparticle level, providing a framework for understanding the dynamic changes of physicochemical properties in nanoparticles during chemical reactions.

Regulatory Science Approach in Pharmaceutical Development of Follow-on Versions of Non-Biological Complex Drug Products

Daniela M. Zagalo et al.

Summary: Scientific and technological breakthroughs in the field of Nanotechnology have played a significant role in the development and approval of Non-Biological Complex Drugs (NBCDs). However, the rapid expansion of NBCDs and their follow-on versions have posed various challenges in terms of science, technology, and regulation. The lack of official recognition and dedicated regulatory pathway for NBCDs, as well as the absence of clear and consistent guidance documents, have hindered the acceptance and potential of these drug products. The inconsistent regulatory approaches across different agencies and countries have also created uncertainty and may affect patient access to high-quality NBCD follow-on versions. Therefore, there is a need to establish a specific regulatory pathway or optimize existing pathways to address the complexity of NBCDs and their follow-on versions. This review aims to critically examine and provide an overview of the current regulatory landscape of NBCDs and follow-on versions, with the goal of filling regulatory and scientific gaps in establishing therapeutic equivalence.
