The PE-PPE Family of Mycobacterium tuberculosis: Proteins in Disguise

Christopher D'Souza et al.

Summary: Mycobacterium tuberculosis has been infecting a third of the world's population and the PE-PPE protein family has been identified as a key factor in its pathogenesis. Despite extensive research, many important questions about these proteins remain unanswered.

Mycobacterium tuberculosis PE/PPE proteins enhance the production of reactive oxygen species and formation of neutrophil extracellular traps

Maria Garcia-Bengoa et al.

Summary: This study found that Mtb proteins PE18, PPE26, and PE31 can stimulate the production of NETs in human blood-derived neutrophils, which is correlated with an increase in intracellular ROS levels. Furthermore, the presence of PMA enhances this effect. Transmission electron microscopy confirmed that neutrophils remain viable and structurally intact after incubation with the PE/PPE proteins, indicating the occurrence of vital NET formation.

Enhanced serodiagnostic potential of a fusion molecule consisting of Rv1793, Rv2628 and a truncated Rv2608 of Mycobacterium tuberculosis

Sadaf Sulman et al.

Summary: The study demonstrated that the fusion protein TriFu64 increased the sensitivity of TB serodiagnosis and was particularly effective in detecting sputum and culture negative patients. By combining Mtb antigens into fusion proteins, a broad sensitivity across different disease types, gender and age groups in a Pakistani population was achieved.

Development and Evaluation of a Fusion Polyprotein Based on HspX and Other Antigen Sequences for the Serodiagnosis of Tuberculosis

Fangbin Zhou et al.

Summary: This study investigated methods to improve the specificity of serological tests for tuberculosis by blocking antibodies and synthesizing peptides, without decreasing sensitivity. A fusion polyprotein displayed higher sensitivity and maintained specificity, demonstrating a potential novel approach for TB diagnostics.
