Genetic Alterations in Members of the Proteasome 26S Subunit, AAA-ATPase (PSMC) Gene Family in the Light of Proteasome Inhibitor Resistance in Multiple Myeloma

Larissa Haertle et al.

Summary: Compared to other neoplasias, Multiple Myeloma is highly responsive to proteasome inhibitors due to its sensitivity to changes in protein homeostasis. Genetic alterations of PSMC genes affect the susceptibility of Multiple Myeloma to proteasome inhibitors, playing a significant role in disease development and resistance. These alterations can serve as biomarkers to predict and monitor patient response, guiding treatment decisions.

19S Proteasome Subunits as Oncogenes and Prognostic Biomarkers in FLT3-Mutated Acute Myeloid Leukemia (AML)

Joshua J. Lara et al.

Summary: PSMD1 and PSMD3 were identified as prognostic biomarkers and potential therapeutic targets in chronic myeloid leukemia and multiple solid tumors. In this study, the expression of 19S proteasome subunits in acute myeloid leukemia patients with FLT3 gene mutations was analyzed. The results showed that high levels of PSMD3 expression were associated with worse overall survival. PSMD3 knockdown impaired colony formation of FLT3+ AML cell lines and increased overall survival in xenograft models. Proteomics analyses suggested a role for PSMD3 in neutrophil degranulation and energy metabolism.

Evaluating the Prognostic and Therapeutic Potentials of the Proteasome 26S Subunit, ATPase (PSMC) Family of Genes in Lung Adenocarcinoma: A Database Mining Approach

Md. Asad Ullah et al.

Summary: This study explores the potential of PSMC family of genes in the diagnosis and treatment of lung adenocarcinoma (LUAD). PSMCs were found to be differentially expressed in LUAD tissues and their expression levels were associated with patient prognosis and cancer stage. PSMCs were also found to be associated with immune cells and modulators in LUAD tissues, suggesting their involvement in LUAD diagnosis and therapeutic interventions. The co-expression of PSMCs with other genes involved in oncogenic processes and metastasis was also observed. Overall, this study suggests that PSMCs could be potential candidates for LUAD diagnostic and therapeutic measures.
