Adipose-derived Mesenchymal Stem Cells Therapy as a new Treatment Option for Diabetes Mellitus

Agnieszka Miklosz et al.

Summary: The increasing prevalence of diabetes mellitus has led to a demand for new therapeutic strategies. Adipose-derived mesenchymal stem cells (ADMSCs) therapy has gained attention due to its therapeutic effects in regulating the immune system and improving insulin sensitivity. However, transplantation of stem cells has associated risks, leading to the exploration of cell-free therapy, such as exosomes or conditioned media, which show even better therapeutic potential. This review summarizes recent outcomes and molecular mechanisms of ADMSCs action in both type 1 and type 2 diabetes treatment, as well as their feasibility, benefits, and limitations.

Using adipose-derived mesenchymal stem cells to fight the metabolic complications of obesity: Where do we stand?

Agnieszka Miklosz et al.

Summary: Obesity is a critical risk factor for metabolic diseases, but adipose-derived mesenchymal stem cells (ADMSCs) have shown potential in improving pancreatic islet cell function and reducing inflammation and oxidative stress in experimental studies. However, there is a lack of clinical studies using ADMSCs for treating metabolic complications, and further research is needed to establish their safety and efficacy in humans.

The Phenotype of the Adipocytes Derived from Subcutaneous and Visceral ADMSCs Is Altered When They Originate from Morbidly Obese Women: Is There a Memory Effect?

Agnieszka Miklosz et al.

Summary: This study investigated the phenotypic and metabolic characteristics of adipocytes derived from ADMSCs in lean and obese postmenopausal women. The results showed that obesity influenced the functionality of progenitor adipocytes, suggesting a memory effect, and there were pronounced changes in the adipocytes derived from different depots.

White adipose tissue dysfunction in obesity and aging

Marjorie Reyes-Farias et al.

Summary: Both obesity and aging are linked to metabolic diseases like type 2 diabetes and cardiovascular disease. Chronic low-grade inflammation of adipose tissue plays a role in the progression of these diseases. Obesity and aging lead to alterations in adipose tissue, triggering a pro-inflammatory phenotype in resident immune cells. They also share characteristics such as a higher visceral fat to subcutaneous fat ratio and a reduced lifespan.

Obesity activates immunomodulating properties of mesenchymal stem cells in adipose tissue with differences between localizations

Camille Lefevre et al.

Summary: The study found that high-fat diet can activate adipose stem cells in subcutaneous and visceral adipose tissue to different degrees early in obesity. These adipose stem cells can inhibit the proliferation of activated T lymphocytes and TNFα expression by macrophages, while releasing higher concentrations of IL6. These differences may contribute to the development of low-grade inflammation in visceral adipose tissue with obesity, while inflammation in subcutaneous adipose tissue is delayed.

Summary: The study found that in an obese mouse model, treatment with AD-MSCs can lower blood glucose levels, improve glucose tolerance, and significantly reduce levels of inflammatory mediators and the atherogenic index of plasma.

Does TBC1D4 (AS160) or TBC1D1 Deficiency Affect the Expression of Fatty Acid Handling Proteins in the Adipocytes Differentiated from Human Adipose-Derived Mesenchymal Stem Cells (ADMSCs) Obtained from Subcutaneous and Visceral Fat Depots?

Agnieszka Miklosz et al.

Summary: TBC1D4 plays a role in regulating fatty acid entry into human adipocytes, with its deficiency leading to the translocation of CD36/SR-B2 transport protein to the plasma membrane, resulting in increased uptake of saturated long-chain fatty acids. The tissue-specific pattern observed, with more pronounced changes in subcutaneous ADMSC-derived adipocytes, suggests a complex regulation mechanism involving TBC1D4 in fatty acid transport.
