Apicobasal transferrin receptor localization and trafficking in brain capillary endothelial cells

Simone S. E. Nielsen et al.

Summary: The mechanisms of transferrin receptor trafficking across brain capillary endothelial cells are not well understood but are important for maintaining brain iron homeostasis and developing drug delivery strategies. This study used expansion microscopy and immunofluorescence-based transport studies to investigate the distribution and sorting of transferrin receptor in these cells. The findings reveal different sorting mechanisms for transferrin receptor depending on the direction of trafficking, with the highest transcytosis capacity observed in the brain-to-blood direction. These results have implications for understanding brain iron homeostasis and optimizing drug delivery to the brain.

Standardized Preclinical In Vitro Blood-Brain Barrier Mouse Assay Validates Endocytosis-Dependent Antibody Transcytosis Using Transferrin-Receptor-Mediated Pathways

Alex Petrovic et al.

Summary: This study developed an in vitro BBB model based on murine cells to evaluate the BBB-penetrating capabilities of transferrin-receptor-targeting antibodies. The results showed that the conjugated antibodies were more effectively transported in the model compared to unconjugated antibodies. The model can be used as a preclinical screening platform for therapeutic neurological pathologies.

A hydrogel model of the human blood-brain barrier using differentiated stem cells

Nandita Rahatekar Singh et al.

Summary: Researchers developed an in vitro model of the human blood-brain barrier using a collagen hydrogel and differentiated endothelial cells from human induced pluripotent stem cells. The model was able to mimic certain characteristics of the blood-brain barrier, but the endothelial-like cells still showed some immature features. The model demonstrated low permeability to certain tracers and functional activity of P-glycoprotein and active transcytosis of polypeptides.

Blood-brain barrier endothelial cells in neurodegenerative diseases: Signals from the barrier

Yiwen Yuan et al.

Summary: As blood-brain barrier (BBB) disruption becomes a common problem in early stages of neurodegenerative diseases, the crucial roles of barrier-type brain endothelial cells (BECs) in the pathophysiology of these diseases are being recognized. The mechanisms of how early vascular dysfunction contributes to neurodegeneration are still unclear, and understanding BEC functions is a promising start. This review presents current evidence of BEC dysfunction in neurodegenerative diseases and explores how BEC signals participate in the pathogenesis of these diseases.

Sortilin regulates blood-brain barrier integrity

Andrea E. Toth et al.

Summary: Sortilin acts as a crucial regulatory protein for maintaining the tightness of the blood-brain barrier (BBB). Lack of sortilin results in BBB leakage, potentially due to alterations in signalling pathways, leading to compromised connections between brain endothelial cells.

Polarized α-synuclein trafficking and transcytosis across brain endothelial cells via Rab7-decorated carriers

Parvez Alam et al.

Summary: In this study, the researchers investigated the uptake and transport mechanisms of alpha-synuclein monomers and oligomers across the blood-brain barrier (BBB). They found that both monomers and oligomers were internalized by brain endothelial cells, with a higher restriction of oligomeric transport. Additionally, they discovered that monomeric alpha-synuclein trafficked via carriers decorated with Rab7. Knockdown of Caveolin1, VPS35, and Rab7 affected the transcytosis of monomeric alpha-synuclein, suggesting a polarized regulation of its vesicular transport. These findings highlight the importance of Rab7 in the trafficking of monomeric alpha-synuclein across the BBB, suggesting it could be a potential target for therapeutic strategies against Parkinson's disease.

Summary: This study investigated the differentiation of human induced pluripotent stem cells (hiPSCs) into brain capillary endothelial-like cells (BCEC) for potential use in drug delivery research. The results demonstrated that the BCEC monolayers exhibited properties of the blood-brain barrier, including barrier tightness, high dynamic range, and functional transport capabilities.

Quantitative correlative microscopy reveals the ultrastructural distribution of endogenous endosomal proteins

Jan van der Beek et al.

Summary: The study investigates the distribution of key endosomal regulators within cells using a quantitative on-section correlative light and electron microscopy approach. Rab5 predominantly marks early endosomes, while EEA1 colocalizes with Rab5 on early endosomes and also labels late endosomes. APPL1 is restricted to small Rab5-positive vesicular profiles, and Rab7 primarily labels late endosomes and lysosomes.

Development of Novel Therapeutics Targeting the Blood-Brain Barrier: From Barrier to Carrier

Jia Li et al.

Summary: The blood-brain barrier is a crucial neurovascular unit that prevents harmful substances from entering the brain and maintains normal neuronal function. Disruption of BBB in diseases can contribute to disease progression, while restoration of BBB integrity is a biomarker for better clinical outcomes. The BBB also plays a role in facilitating communication between neuronal and circulatory systems, showing promising results for brain-targeted drug delivery.
