Summary: Alternaria black spot disease, caused by Alternaria alternata, is a serious threat to the pecan industry in South Africa and global. Genetic diversity of A. alternata isolates from different locations in South Africa was investigated. A rapid and reliable technique using PCR-RFLP analysis and two endonucleases was developed for screening the pathogen causing Alternaria black spot disease.

Multigene Phylogeny and Pathogenicity Trials Revealed Alternaria alternata as the Causal Agent of Black Spot Disease and Seedling Wilt of Pecan (Carya illinoinensis) in South Africa

Summary: The study aimed to identify the causative agents of Alternaria black spot and seedling wilt in South African pecan orchards. Molecular techniques were used to retrieve thirty Alternaria isolates from symptomatic and non-symptomatic pecan plant organs. The isolates were all members of Alternaria alternata sensu stricto and showed pathogenicity in detached nuts and leaves, confirming that A. alternata causes black spot disease and seedling wilt of pecans. This study provides one of the first documentations of Alternaria black spot disease in South African pecan trees.

Phenotypic and molecular characterization of the resistance to azoxystrobin and pyraclostrobin in Fusarium graminearum populations from Brazil

Summary: The study characterized 225 strains of F. graminearum from two states in Brazil in regards to their response to two QoI fungicides. The median EC50 values for azoxystrobin and pyraclostrobin varied between the two populations, with no evidence of cross-resistance detected. The frequency of less-sensitive isolates increased over time in the RS state of Brazil.

CYP51 Mutations in the Fusarium solani Species Complex: First Clue to Understand the Low Susceptibility to Azoles of the Genus Fusarium

Summary: This study investigated the potential relationship between azole susceptibility and mutations in CYP51 protein sequences in the Fusarium solani species complex (FSSC). The results showed point mutations in FSSC similar to those described in azole-resistant strains. Protein sequence alignments of CYP51 revealed different patterns, and a link between voriconazole susceptibility and protein sequences was observed.

Summary: This study investigated the impact of cyp51 genes on voriconazole response and resistance in Aspergillus niger and Aspergillus tubingensis. It was found that cyp51A is more actively involved in the transcriptional response to azole stress compared to cyp51B, while both enzymes similarly impact the total ergosterol content within the cell. These findings contribute to a better understanding of Aspergillus azole resistance, especially in non-A. fumigatus species.

Distribution of Alternaria alternata isolates with resistance to quinone outside inhibitor (QoI) fungicides in Brazilian orchards of tangerines and their hybrids

Summary: This study investigated the sensitivity and resistance distribution of A. alternata to QoI fungicides in citrus trees cultivated in different regions of Sao Paulo. The results showed that in several municipalities, 100% of isolates were resistant to both fungicides, suggesting the need for anti-resistance strategies in disease management.

Characterization of the Molecular Mechanisms of Resistance against DMI Fungicides in Cercospora beticola Populations from the Czech Republic

Summary: Cercospora leaf spot (CLS) caused by Cercospora beticola is a significant foliar pathogen of sugar beet worldwide. The study assessed DMI resistance in Czech C. beticola isolates and identified a Y464S mutation in the Cyp51 gene as a likely factor contributing to resistance against DMI fungicides. Induced Cyp51 gene expression after fungicide treatment was also observed, supporting the conclusion that gene overexpression may play a role in DMI resistance.
