Development of Efficient Genome-Reduction Tool Based on Cre/loxP System in Rhodococcus erythropolis

Summary: This study developed an effective plasmid-curing method and a simple two-step genome-reduction method using a modified Cre/loxP system. A mutant strain that cured all four plasmids and deleted seven chromosomal regions was successfully obtained in this study. This genetic tool will help develop and improve Rhodococcus strains for various industrial and environmental applications.

Optimization of carotenoid production by Umbelopsis ramanniana

Summary: In this study, Umbelopsis ramanniana was investigated to enhance carotenoid production. Various carbon and nitrogen sources were evaluated, with KNO3 and lactose being the most effective nitrogen and carbon sources, respectively. The optimization of medium components was achieved using Plackett-Burman design, and Box-Behnken response surface methodology was applied to further optimize carotenoid and biomass production. The optimum conditions for maximum carotenoid and biomass production were determined as a lactose concentration of 32.42 g/L, a carbon to nitrogen ratio of 20:1, and a shaking speed of 130 rpm, resulting in a significant increase in carotenoid and biomass production.

Biodiesel production by oleaginous bacteria Rhodococcus opacus PD630 using waste paper hydrolysate

Summary: The utilization of waste paper for biodiesel production has environmental and economic advantages. In this study, waste paper hydrolysate (WPH) was used as a substrate for microbial lipid production. The medium with NH(4)C1 as nitrogen source yielded high biomass and lipid content. Under optimized conditions, the maximum biomass, lipid yield, and lipid content obtained were 6.51 g/L, 2.8 g/L, and 43% respectively. Extracted lipids showed similarities to vegetable oils, and the biodiesel properties were within the permissible limits of international standards.

Effect of process mode, nitrogen source and temperature on L-malic acid production with Aspergillus oryzae DSM 1863 using acetate as carbon source
