Combination Effect of Engineered Endolysin EC340 With Antibiotics

Hye-Won Hong et al.

Summary: This study focused on engineering bacteriophage lysins for improved activity against Gram-negative bacteria and enhanced permeability through the outer membrane. Testing the engineered lysin in combination with standard antibiotics, a synergistic effect was observed with colistin and an additive effect was seen with several other antibiotics. However, there was no generalized pattern in terms of antibiotic class or mechanism of action.

Mycobacteriophages: From Petri dish to patient

Graham F. Hatfull

Summary: Mycobacteriophages play a significant role in understanding viral diversity and evolution. They provide resources and tools for advancing Mycobacterium genetics and show potential in therapeutic applications. Experimental studies with the model organism Mycobacterium smegmatis have provided high-resolution insights into the phages that infect common host strains.

Global Tuberculosis Report 2020-Reflections on the Global TB burden, treatment and prevention efforts

Jeremiah Chakaya et al.

Summary: The global TB report for October 2020 highlights slow progress in TB control efforts and the need to ramp up screening and diagnostic test services. Key drivers of TB, including undernutrition, poverty, diabetes, tobacco smoking, and household air pollution, must be addressed to achieve WHO 2035 TB care and prevention targets. Global focus is needed on the development of new vaccines for TB using new technological methods.

Mycobacteriophages to Treat Tuberculosis: Dream or Delusion?

Andreas H. Diacon et al.

Summary: Global antimicrobial resistance rates are increasing while new antibiotics are in short supply, posing risks for patients with diseases caused by drug-resistant bacteria. Bacteriophages are viruses that attack bacteria and can be used to prevent contamination by human pathogens.

Toward a Phage Cocktail for Tuberculosis: Susceptibility and Tuberculocidal Action of Mycobacteriophages against Diverse Mycobacterium tuberculosis Strains

Carlos A. Guerrero-Bustamante et al.

Summary: Research suggests using bacteriophages may be a promising strategy for treating tuberculosis, with a five-phage cocktail effectively killing Mycobacterium tuberculosis strains without affecting antibiotic effectiveness. This offers new possibilities for treating tuberculosis and combating antibiotic resistance.

PLAN-M; Mycobacteriophage Endolysins Fused to Biodegradable Nanobeads Mitigate Mycobacterial Growth in Liquid and on Surfaces

Courtney G. Davies et al.

Summary: The study demonstrates that Mycobacteriophage endolysins fused to biodegradable polyhydroxyalkanoate (PHA) nanobeads can effectively lyse M. smegmatis MC2 155 cells, achieving up to 79% population reduction. These nanobeads show potential as a protective layer on textiles, with Lysin B enzymes outperforming Lysin A enzymes in surface assays.

Exploration of Synergistic Action of Cell Wall-Degrading Enzymes against Mycobacterium tuberculosis

Loes van Schie et al.

Summary: The study explores the potential effectiveness of cell wall-degrading enzymes in inhibiting Mycobacterium tuberculosis, revealing that enzymes like LysB and lysozymes show potency as inhibitors, especially in the presence of surfactant. The synergistic combination of LysB, alpha-amylase, and polysorbate 80 proved to be the most effective in disrupting the mycolic acid barrier, suggesting potential for the development of tailored antimycobacterial enzymes. Delivery of such therapeutic enzymes through airway administration has been successful and merits further investigation in animal models of active TB.
