Culture of Piglet Intestinal 3D Organoids from Cryopreserved Epithelial Crypts and Establishment of Cell Monolayers

Eloise Mussard et al.

Summary: This article describes a method for culturing pig intestinal 3D organoids using cryopreserved epithelial crypts. The advantages of this method include the temporal dissociation of crypt isolation and organoid culture, the ability to obtain large stocks of cryopreserved crypts from multiple animals and segments of the intestine, and the reduction in the need for fresh tissue sampling from live animals. Additionally, a protocol for establishing cell monolayers derived from 3D organoids is detailed to study interactions with nutrients, microbes, or drugs at the apical side of the epithelial cells.

Review: Can early-life establishment of the piglet intestinal microbiota influence production outcomes?

T. L. Nowland et al.

Summary: The gastrointestinal tract microbiota plays a crucial role in the development and function of various body processes. The early-life microbial colonisation has a significant impact on intestinal and immune development, and disturbances during this period can have long-lasting negative effects on the host. Therefore, understanding how to improve initial intestinal microbiota colonisation in piglets is essential for pork production.

Dynamics of the Gut Microbiome in Shigella-Infected Children during the First Two Years of Life

Esther Ndungo et al.

Summary: Shigella continues to be a major contributor to diarrheal illness and dysentery in children under 5 in low- and middle-income countries. This study explored the impact of Shigella infection on the developing gastrointestinal microbiome in infants and found temporal changes in the microbiota associated with the infection. These findings highlight the need for further research on the microbial communities associated with disease susceptibility and resolution.

The phenotype of the gut region is more stably retained than developmental stage in piglet intestinal organoids

Eloise Mussard et al.

Summary: Intestinal organoids generated from piglets retained a location-specific phenotype but not a developmental stage-specific phenotype. The luminal environment of the organoids was not representative of the in vivo environment, suggesting that reproducing the luminal environment might improve the physiological relevance of intestinal organoids.

Longitudinal Study of Fecal Microbiota in Calves with or without Diarrhea Episodes before Weaning

Pau Obregon-Gutierrez et al.

Summary: By analyzing the changes in the fecal microbiota of calves, differences related to their health status were found. The microbiota in healthy animals stabilized earlier and showed changes in low abundant bacteria that may play a role in their subsequent health. On the other hand, specific bacteria were found in the fecal microbiota of calves with diarrhea, indicating a higher predisposition to suffer from diarrhea. These findings can contribute to the development of new probiotics for promoting gut health in calves.

Gut Microbiota-Derived Metabolite Signature in Suckling and Weaned Piglets

Martin Beaumont et al.

Summary: The gut microbiota produces different metabolites in suckling and weaned piglets, including short-chain fatty acids and biogenic amines. The origin of the piglets also impacts the levels of certain bacterial metabolites. The fecal metabolome signatures in suckling and weaned piglets are associated with specific microbiota-predicted functionalities, structure, and diversity.

Early life feeding accelerates gut microbiome maturation and suppresses acute post-weaning stress in piglets

R. Choudhury et al.

Summary: The study revealed that early feeding has a positive impact on the microbiota development in neonatal piglets, promoting microbial maturation and benefiting piglet health and performance during the transition period to weaning.

How biological sex of the host shapes its gut microbiota

Francesco Valeri et al.

Summary: The gut microbiota is a complex system that is influenced by various factors, including sex. Despite limited understanding of the mechanisms behind sex-related differences, it is suggested that sex hormones, dietary habits, and antibiotics may have a direct impact on the composition of the gut microbiota. More research is needed to fully grasp the role of sexual hormones, particularly testosterone, in shaping the gut microbiota.

Development of piglet gut microbiota at the time of weaning influences development of postweaning diarrhea - A field study

Daniela Karasova et al.

Summary: The study revealed that certain microbial populations before weaning may increase the risk of piglets developing diarrhea, while other microbial populations after weaning may enhance piglet health and disease resistance. Therefore, changes in microbiota before and after weaning are crucial for piglet resistance to postweaning diarrhea under field conditions.

Microbial Regulation of Host Physiology by Short-chain Fatty Acids

Bart van der Hee et al.

Summary: The ancestral diet had more fiber, while modern society's diet lacks it. SCFAs produced by gut bacteria fermenting fiber play a crucial role in intestinal homeostasis and the pathophysiology of human diseases. SCFAs can also impact tissues and organs beyond the gut through blood circulation, inducing epigenetic changes in the genome.

The fecal microbiota of piglets during weaning transition and its association with piglet growth across various farm environments

Diana Luise et al.

Summary: This study investigates the fecal microbiota of piglets in different living environments during the weaning transition and identifies characteristics associated with good growth of piglets after weaning. The study reveals the impact of rearing environment on microbial composition and development, as well as the presence of ubiquitous enterotypes among different farms reflecting maturational stages of microbiota.
