Solid-phase microextraction of endogenous metabolites from intact tissue validated using a Biocrates standard reference method kit

Runshan Will Jiang et al.

Summary: Improved analytical methods using solid-phase microextraction (SPME) fibers were utilized in this study to monitor changes in endogenous compounds in homogenized and intact ovine lung tissue. The findings highlight the importance of sample preparation in tissue-based metabolomics studies and the unique ability of SPME to perform minimally invasive extractions while providing broad metabolite coverage.

Eight Zhes Decoction ameliorates the lipid dysfunction of nonalcoholic fatty liver disease using integrated lipidomics, network pharmacology and pharmacokinetics

Yuping Zhou et al.

Summary: This study used a strategy integrating lipidomics, network pharmacology, and pharmacokinetics to reveal the active components and mechanisms of Eight Zhes Decoction (EZD) against nonalcoholic fatty liver disease (NAFLD). The results showed that EZD attenuated collagen deposition and steatosis in the livers of NAFLD model mice. Naringenin, artemetin, canadine, and bicuculline were identified as the active ingredients of EZD against NAFLD.

Erythrocyte sphingolipid species as biomarkers of Alzheimer's disease

Jericha Mill et al.

Summary: Diagnosing Alzheimer's disease in the early stage is challenging. This study explores the potential utility of erythrocyte metabolite profiles as biomarkers for screening AD. Comparing the metabolomes of AD patients and control group, increased amino acid metabolism and decreased lipid metabolism were observed in AD. Sphingolipids, previously implicated in AD, were consistently decreased in AD patients, suggesting their potential as risk markers for AD development.

Novel Strategy for Mining and Identification of Acylcarnitines Using Data-Independent-Acquisition-Based Retention Time Prediction Modeling and Pseudo-Characteristic Fragmentation Ion Matching

Chao Feng et al.

Summary: A new method based on retention time prediction and data-independent acquisition strategies was proposed for the large-scale identification of acylcarnitines. The introduction of relative cumulative eluotropic strength as a novel descriptor solves the problem of accurately predicting acylcarnitines with long carbon chains in traditional models. The method effectively reduces false-positive results by providing access to predictive RT and MS2 spectra of suspect features.

Effect of Microbial Status on Hepatic Odd-Chain Fatty Acids Is Diet-Dependent

Karolin Weitkunat et al.

Summary: The study shows that odd-chain fatty acids (OCFA) are associated with obesity and type-2-diabetes, and their formation is influenced by diet and the microbiota. Bacterial propionate production affects hepatic OCFA formation, but this effect may be masked by dietary propionate intake.

Absolute Quantification of Acylcarnitines Using Integrated Tmt-PP Derivatization-Based LC-MS/MS and Quantitative Analysis of Multi-Components by a Single Marker Strategy

Tian Tang et al.

Summary: This study utilized microwave-assisted Tmt-PP derivatization and the QAMS strategy to improve the accuracy of LC-MS/MS determination of ACs, reducing reaction time and resolving the standard availability issue. The established workflow was applied to discover dysregulated ACs in xenograft colon cancer mice, with quantification results highly comparable to traditional methods.
