Innate immune cell-intrinsic ketogenesis is dispensable for organismal metabolism and age-related inflammation

Summary: Aging is accompanied by chronic low-grade inflammation, and ketone bodies have been found to have potential in improving healthspan in aging mouse models. The most abundant ketone body, beta-hydroxybutyrate, inhibits the NLRP3 inflammasome in myeloid cells, suggesting it may serve as a metabolic checkpoint for inflammation. By disrupting the expression of the enzyme required for ketogenesis, the researchers found that liver-derived circulating ketone bodies may play a crucial role in deactivating the NLRP3 inflammasome and controlling organismal metabolism during aging.

This review summarizes the functions of insulin in glucose metabolism and discusses the mechanisms proposed for insulin resistance, such as ectopic lipid accumulation, endoplasmic reticulum stress, and inflammation. Potential therapeutic strategies for addressing insulin resistance are also suggested.

An explanation for negligible senescence in animals

Summary: Negligible or negative senescence, where mortality risk remains stable or decreases with age, has been observed in some wild animals. However, the reason why there is no evidence of senescence in these animals is not fully understood. Our research reveals that the reported lifespans of wild animals with low recovery probability significantly underestimate the increase in mortality risk, leading to the appearance of negligible senescence. This finding provides an explanation for the phenomenon of negligible senescence in many wild animals.

Summary: Using a statistical approach, the study forecasts the maximum reported age at death (MRAD) through 2100. The findings suggest a greater than 99% probability that the current MRAD of 122 will be surpassed by 2100, with probabilities of reaching at least 126, 128, or 130 years old this century estimated at 89%, 44%, and 13% respectively.

Summary: Patients with T2DM and/or CVD have lower levels of osteocalcin and bone remodelling, leading to increased risk of fragility fractures. Hyperinsulinaemia in these patients is associated with normal to increased bone mineral density and higher risk of fragility fractures. Hyperinsulinaemia leads to decreased antioxidant activity and mitochondrial dysfunction, affecting osteoblast/cytogenesis and osteocyte viability.
