Andrographolide induced ferroptosis in multiple myeloma cells by regulating the P38/Nrf2/HO-1 pathway

Wenxia Li et al.

Summary: Andrographis paniculata, a traditional Chinese medicine, has been used as a functional food in Asia. Andrographolide, a compound isolated from Andrographis paniculata, has potent anticancer activity. In this study, researchers found that Andro induced cell death in multiple myeloma (MM) cells through a type of cell death called ferroptosis, and this mechanism may provide a potential preventative and therapeutic approach for MM.

Crosstalk between Yeast Cell Plasma Membrane Ergosterol Content and Cell Wall Stiffness under Acetic Acid Stress Involving Pdr18

Ricardo A. Ribeiro et al.

Summary: This study investigates the impact of Pdr18 expression on yeast cell wall during adaptation to acetic acid stress. Results show that Pdr18 deletion leads to changes in transcriptional activation of cell wall biosynthetic genes and cell wall stiffness and polysaccharide content. Despite a stronger and more long-lasting adaptive response, the pdr18 Delta population has lower resistance to lyticase activity and requires a longer period for intracellular pH recovery and growth resumption. Plasma membrane ergosterol content is drastically reduced under acetic acid stress, especially in pdr18 Delta cells, suggesting a coordinated response between plasma membrane and cell wall against the detrimental effects of acetic acid.

Potential Role of the Host-Derived Cell-Wall Binding Domain o Endolysin CD16/50L as a Molecular Anchor in Preservation of Uninfected Clostridioides difficile for New Rounds of Phage Infection

Wichuda Phothichaisri et al.

Summary: This study investigates the function of the cell-wall binding domain (CBD) of an endolysin from a C. difficile phage. The CBD is found to play a role in anchoring the enzyme to post-lytic cell-wall remnants, limiting enzyme diffusion and preserving neighboring host cells, which facilitates phage replication. This insight into the regulation of endolysin through CBD may have potential applications in treating C. difficile infection.

Triazole Derivatives Target 14α-Demethylase (LDM) Enzyme in Candida albicans Causing Ergosterol Biosynthesis Inhibition

Irfan A. Rather et al.

Summary: Candida albicans is a common cause of fungal infections in humans and azoles are commonly used for treatment. However, their overuse has led to drug resistance. In this study, new triazole-based molecules were developed and tested against Candida albicans. In vitro and in silico results showed that these derivatives have the potential to be effective antifungal drugs, as they effectively bind to the target enzyme, inhibit ergosterol production, and prevent biofilm formation.

Receptor binding protein of prophage reversibly recognizes the low-molecular weight subunit of the surface-layer protein SlpA in Clostridioides difficile

Tanaporn Phetruen et al.

Summary: In this study, the receptor-binding protein (RBP) Pts(HN10)M was identified and shown to play a crucial role in mediating the interaction between phage and C. difficile cells. The C-terminal domain of Pts(HN10)M and the low-molecular weight subunit of surface-layer protein A (SlpA) were found to be important for the binding process. This research highlights the potential of Pts(HN10)M as an RBP for phage engineering and therapy against C. difficile infection.
