Validation and Functional Analysis of Reference and Tissue-Specific Genes in Adipose Tissue of Freshwater Drum, Aplodinotus grunniens, under Starvation and Hypothermia Stress

Summary: In this study, reference genes and tissue-specific genes were identified in the adipose tissue of Aplodinotus grunniens, and their function under exogenous starvation and hypothermic stress was explored. Results showed that 60SRP was the most stable reference gene in adipose tissue. In addition, seven tissue-specific genes were confirmed in adipose tissue and were found to be active in response to starvation and hypothermic stress in a time- or temperature-dependent manner. These findings provide tissue-specific targets for adipose regulation in A. grunniens.

Gut Microbes Reveal Pseudomonas Medicates Ingestion Preference via Protein Utilization and Cellular Homeostasis Under Feed Domestication in Freshwater Drum, Aplodinotus grunniens

Summary: This study conducted a 4-month feeding domestication experiment with freshwater drum larvae to evaluate the underlying mechanism and molecular targets responsible for alterations in ingestion performance. The results showed that feeding domestication significantly increased body weight, digestive enzyme activity, and activated genes related to appetite and perception. Microbiome and host transcriptome analysis revealed that Pseudomonas-mediated ingestion capacity, protein utilization, and cellular homeostasis might be the underlying mechanism for feed domestication.

Summary: Compared with other species, freshwater fish have a more efficient biosynthetic pathway for synthesizing DHA. This study investigated the role of Ehhadh in the synthesis of DHA in zebrafish. The results showed that knockout of Ehhadh inhibited DHA synthesis, while overexpression of Ehhadh increased DHA content in the liver. Additionally, Ehhadh was found to affect the expression of genes related to PUFA synthesis and SREBF and PPAR signaling pathways.

Summary: The acute exposure of swordtail fish to microplastics was found to have adverse effects on their liver, leading to changes in metabolic profiles involving multiple metabolic pathways and metabolites. Microplastics affected oxidative stress, immune function, energy metabolism, sugar metabolism, lipid metabolism, and other physiological processes, as well as feed utilization, growth performance, nutrient metabolism, and animal growth. Additionally, a dose-effect relationship was observed between the quantity of microplastics and the number of affected amino acids.

Hypothermia-Mediated Apoptosis and Inflammation Contribute to Antioxidant and Immune Adaption in Freshwater Drum, Aplodinotus grunniens

Summary: This study investigates the effects of hypothermia on the freshwater drum and finds that it induces oxidative stress and immunosuppression. The ultrastructure of the liver also undergoes changes under hypothermia exposure. Transcriptome analysis reveals differentially expressed genes involved in metabolism, immunity, inflammation, programmed cell death, and disease. In addition, hypothermia induces inflammatory response and apoptosis in different immune organs, which are correlated with antioxidation and immunity suppression. Overall, these findings enhance our understanding of how the freshwater drum adapts to hypothermia stress.

Acetylferulic paeonol ester: A new feed additive reduces lipid accumulation in the liver of Nile tilapia (Oreochromis niloticus) by modulating lipid and glucose metabolism

Summary: This study investigated the effects of different levels of acetylferulic paeonol ester (APE) on the growth performance, body composition, serum biochemical indicators, and lipid and glucose metabolism of Nile tilapia. The results showed that APE supplementation improved weight gain and decreased liver lipid content. It also affected serum biochemical parameters related to lipid and glucose metabolism. Transcriptome analysis revealed changes in gene expression related to fatty acid oxidation and degradation. APE influenced the hepatic glucose and lipid metabolism of tilapia at both enzymatic and mRNA levels.

Summary: Fatty liver commonly occurs in intensively farmed tilapia, leading to impairments in hepatic lipid metabolism and disruptions to protein and carbohydrate metabolism. In this study, high-fat diet fed tilapia accumulated more fat in the liver compared to normal-fat diet fed tilapia, with differential accumulation of 36 metabolites between the two groups. These metabolic disorders may be related to the lower growth rate observed in high-fat diet fed tilapia.
