Semi-synthetic terpenoids with differential adjuvant properties as sustainable replacements for shark squalene in vaccine emulsions

Karl J. J. Fisher et al.

Summary: Synthetic biology has been used to produce supply-limited sesquiterpenoids, such as artemisinin and beta-farnesene. As the overfishing of deep-sea sharks has led to potential supply problems for squalene, a triterpenoid derived from shark liver oil, researchers have chemically generated squalene analogues from fermentation-derived beta-farnesene to serve as sustainable vaccine adjuvants. By evaluating the adjuvant activity of these analogues, they have identified compounds with enhanced, equivalent, or decreased activity compared to shark squalene emulsion, shedding light on the structural properties important for adjuvant activity.

Macrophage-like THP-1 Cells Derived from High-Density Cell Culture Are Resistant to TRAIL-Induced Cell Death via Down-Regulation of Death-Receptors DR4 and DR5

Yana Vladimirovna Lomovskaya et al.

Summary: The resistance of leukemic cells to TRAIL-induced apoptosis is a major challenge in the treatment of leukemia. In this study, researchers discovered that human acute myeloid leukemia cells, when subjected to high-density culture conditions, displayed a macrophage-like phenotype and increased resistance to TRAIL-induced cell death. This resistance can be attributed to a decrease in the expression of death receptors DR4 and DR5 on the leukemic cells. The findings suggest that stress conditions in high-density cell cultures may contribute to tumor progression by promoting TRAIL resistance.

Summary: This study developed robust Saccharomyces cerevisiae strains with enhanced squalene accumulation through metabolic engineering and adaptive evolution, identifying the F420I mutation of ERG1 as beneficial for squalene synthesis. Increased squalene was found to result in higher neutral lipid content and clustering of lipid droplets in the evolved mutant strains.

Summary: Flavonoids containing a catechol moiety exhibit higher antioxidant activity, while products formed by their condensation with toxic carbonyl compounds show high antioxidant properties. It is important to note that the conditions under which parent flavonoids and their products exhibit maximal antioxidant activity may differ.
