Targeting the Androgen Signaling Axis in Prostate Cancer

Charles Dai et al.

Summary: The activation of androgen receptor (AR) and AR-driven transcriptional programs play critical roles in the pathophysiology of prostate cancer. However, therapeutic resistance often occurs due to molecular alterations in the androgen signaling axis. The efficacy of next-generation AR-directed therapies provides validation for the dependence on AR signaling, but metastatic prostate cancer remains incurable. Therefore, it is important to understand the diverse mechanisms by which tumors resist AR-directed therapies to find new treatment options.

Cancer statistics, 2022

Rebecca L. Siegel et al.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

G9a inhibition by CM-272: Developing a novel anti-tumoral strategy for castration-resistant prostate cancer using 2D and 3D in vitro models

Filipa Moreira-Silva et al.

Summary: In this study, the researchers found that CM-272 inhibits the viability and proliferation of prostate cancer cells, making it a potential anti-tumor drug. They also discovered that CM-272 specifically inhibits the activity of G9a, while not significantly affecting DNMT1 activity. The mechanism of action of CM-272 appears to be dependent on the catalytic function of G9a. Additionally, CM-272 demonstrates anti-tumor effects in 3D prostate cancer models.

From Therapy Resistance to Targeted Therapies in Prostate Cancer

Filipa Moreira-Silva et al.

Summary: Prostate cancer is the second most common malignancy among men. Early-stage disease is curable, but advanced stage PCa is mostly incurable and resistant to standard therapeutic options. Individualized analysis and selective targeted therapy may improve treatment outcomes.

Epigenetically-regulated miR-30a/c-5p directly target TWF1 and hamper ccRCC cell aggressiveness

Goncalo Outeiro-pinho et al.

Summary: MiR-30a/c-5p is downregulated and associated with aggressiveness in ccRCC. Downregulation of miR-30a/c-5p is accompanied by increased promoter methylation, which can be restored by decitabine treatment. MiR-30a/c-5p not only decreases cell aggressiveness, but also upregulates epithelial markers and downregulates EMT-promoting players.

Summary: Targeting the androgen receptor signaling axis has long been the main strategy for prostate cancer therapy, but stronger inhibition has led to a subset of cancers becoming AR-negative, with a poorer prognosis.

Single cell epigenetic visualization assay

Sam Kint et al.

Summary: Characterization of individual cells' epigenetic status remains challenging due to limited coverage of current sequencing approaches. A targeted microscopy-based epigenetic visualization assay (EVA) was developed to overcome these limitations, allowing for detection and quantification of epigenetic marks at genes of interest in single cells. By linking epigenetic marks to gene transcription, the efficacy of the method was validated.

Cytostatic Action of Novel Histone Deacetylase Inhibitors in Androgen Receptor-Null Prostate Cancer Cells

Zohaib Rana et al.

Summary: Jazz90 and Jazz167 act as cytostatic HDAC inhibitors in AR-null prostate cancer cells by inhibiting cell growth with selective cytotoxicity, reducing HDAC activity, and increasing acetylation levels in cells. However, their apoptotic effects on PC3 and DU145 cells are limited, showing only a maximum of 7% cell population undergoing apoptosis after treatment with the compounds.
