Technical issues still remain in gene therapy, especially in the clinical use of viral vectors. Non-viral gene transfection agents are emerging as potentially safe and effective alternatives for treating genetic and acquired diseases.

High cholesterol absorption: A risk factor of atherosclerotic cardiovascular diseases?

Summary: This review discusses the relationship between cholesterol metabolism and the risk reduction of atherosclerotic cardiovascular diseases (ASCVDs). It focuses on the potential mechanisms of high cholesterol absorption and its association with ASCVDs. Loss-of-function genetic variations in ABCG5, ABCG8, and NPC1L1 have been found to affect cholesterol absorption and are associated with the risk of ASCVDs. Combination therapy of statin with cholesterol absorption inhibition is necessary for individuals with high cholesterol absorption to reduce ASCVD risk.
