The CYTOLD and ERTOLD pathways for lipid droplet-protein targeting

Maria-Jesus Olarte et al.

Summary: In this review, the two pathways for direct LD protein localization are discussed: the CYTOLD pathway mediates protein targeting from the cytosol to LDs, and the ERTOLD pathway functions in protein targeting from the endoplasmic reticulum to LDs. The emerging principles for each targeting pathway in animal cells are described, along with highlighting open questions in the field.

The surface of lipid droplets constitutes a barrier for endoplasmic reticulum-resident integral membrane proteins

Rasha Khaddaj et al.

Summary: This study investigates the localization of integral membrane proteins to the surface of lipid droplets (LDs) and confirms that the ER-LD junction acts as a barrier for ER-resident integral membrane proteins. The study also reveals that certain membrane proteins can be targeted to LDs through LD-targeted proteins.

Seipin collaborates with the ER membrane to control the sites of lipid droplet formation

Roger Schneiter et al.

Summary: Most cells store metabolic energy in lipid droplets (LDs), which consist of a hydrophobic core covered by a phospholipid monolayer. Recent evidence suggests that LD formation occurs in specific subdomains of the endoplasmic reticulum (ER) where proteins gather to initiate LD assembly. The lipodystrophy protein Seipin plays a crucial role in LD biogenesis by forming a large oligomeric toroidal structure that promotes LD nucleation. This article discusses the role of LD biogenesis factors that interact with Seipin to assemble functional LDs.

Lipid droplets form a network interconnected by the endoplasmic reticulum through which their proteins equilibrate

Stephanie Cottier et al.

Summary: Lipid droplets (LDs) are intracellular structures for neutral lipid storage, and their protein transfer is influenced by yeast cell fusion and the ER membrane. This transfer is continuous, bidirectional, and the LDs are interconnected through the ER membrane.

Seipin accumulates and traps diacylglycerols and triglycerides in its ring-like structure

Valeria Zoni et al.

Summary: Lipid droplets are intracellular organelles responsible for lipid storage that emerge from the endoplasmic reticulum. Through molecular dynamics simulations, it has been revealed that seipin promotes lipid droplet formation by clustering lipids in its unconventional ring-like oligomeric structure, suggesting a role in priming specific ER sites for LD biogenesis.

Mechanism of lipid droplet formation by the yeast Sei1/Ldb16 Seipin complex

Yoel A. Klug et al.

Summary: The study revealed the mechanism of lipid droplet formation by the coordination of yeast Seipin Sei1 and its partner Ldb16. Sei1 assembles a homooligomeric ring in the ER to concentrate triacylglycerol, promoting Ldb16 stability and lipid droplet assembly.

A Unique Junctional Interface at Contact Sites Between the Endoplasmic Reticulum and Lipid Droplets

Vineet Choudhary et al.

Summary: Lipid droplets (LDs) store metabolic energy in the form of neutral lipids and originate from the endoplasmic reticulum (ER), maintaining contact with it to facilitate protein and lipid exchange. Proper formation of ER-LD junctions is crucial to prevent the formation of aberrant LDs, which can impact lipid homeostasis and lead to lipid storage diseases. The close contacts between LDs, peroxisomes, and mitochondria play a role in metabolic channeling of fatty acids, highlighting the importance of LD contact sites in lipid metabolism.

Ultrastructural plasma membrane asymmetries in tension and curvature promote yeast cell fusion

Olivia Muriel et al.

Summary: In the process of cell-cell fusion in walled fission yeast, researchers have discovered marked asymmetries between gametes, with the plasma membrane of h- cells protruding into h+ cells. This asymmetry is relaxed upon fusion, with the formation of ramified fusion pores. This suggests that differential turgor pressure and exocytosis/endocytosis ratios between mating types favor cell-cell fusion.
