Identification of tumor antigens with immunopeptidomics

Chloe Chong et al.

Summary: Identification of actionable tumor antigens is crucial for various cancer immunotherapies, including T cell receptor-transduced T cells and patient-specific mRNA or peptide vaccines. By eluting human leukocyte antigen-bound peptides from tumors and subjecting these to mass spectrometry analysis, noncanonical antigens can be discovered, which may lead to more effective treatment options.

Identification of shared tumor epitopes from endogenous retroviruses inducing high-avidity cytotoxic T cells for cancer immunotherapy

Paola Bonaventura et al.

Summary: Researchers identified shared CD8(+) T cell epitopes derived from cancer-associated human endogenous retroviruses (HERVs) in solid tumors using a bioinformatic approach. These epitopes were shown to be immunogenic and capable of recognizing and killing tumor cells expressing the corresponding epitopes, highlighting their potential for the development of cancer vaccines or T cell-based immunotherapies.

Endogenous Retroviral-K Envelope Is a Novel Tumor Antigen and Prognostic Indicator of Renal Cell Carcinoma

Veronika Weyerer et al.

Summary: The study found that ERV-K env protein expression was highest in ccRCC, with the strongest significant membrane expression compared to other RCC subtypes. High ERV-K env total protein expression of all tumor subtypes significantly correlated with low tumor grading and a longer disease-specific survival.

CD8+ T-cell Immune Surveillance against a Tumor Antigen Encoded by the Oncogenic Long Noncoding RNA PVT1

Summary: The study found enriched cryptic HLA-I class ligands in human colorectal cancer tissues, including peptides derived from long noncoding RNA, suggesting that cancer patient T cells are capable of sensing aberrations in noncoding regions of the genome.

Proteogenomic identification of an immunogenic HLA class I neoantigen in mismatch repair deficient colorectal cancer tissue

Tomomi Hirama et al.

Summary: The study identified a rare neoantigen in mismatch repair-deficient colorectal cancer tissue, which was associated with abundant tumor-infiltrating lymphocytes and an inflamed gene expression status. CD8(+) TILs predominantly recognized the detected neoantigen and exhibited functional and specific responses.
