Fragment-to-Lead Medicinal Chemistry Publications in 2021

LSD1 inhibitors for cancer treatment: Focus on multi-target agents and compounds in clinical trials

Summary: Histone lysine-specific demethylase 1 (LSD1/KDM1A) is an epigenetic enzyme that demethylates specific lysine residues of histone H3. Its overexpression is associated with various cancers and leads to cellular-level effects such as differentiation block and increased proliferation, migration, and invasiveness. There are covalent and non-covalent LSD1 inhibitors, some of which are currently in clinical trials for cancer treatment. This review summarizes the structure and functions of LSD1, its pathological implications, and the development of LSD1 inhibitors as potential anticancer agents.

Lysine-Specific Demethylase 1 Promises to Be a Novel Target in Cancer Drug Resistance: Therapeutic Implications

Summary: Chemotherapy, targeted therapy, and immunotherapy can effectively treat most tumors, but drug resistance limits their success. Lysine-specific demethylase 1 (LSD1) plays a role in regulating tumor cell invasion, metastasis, apoptosis, and immune evasion, which are associated with tumor development and progression. Recent studies have found that LSD1 inhibition can restore sensitivity of tumor cells to anticancer drugs containing immune checkpoint inhibitors (ICIs) through various pathways. This review discusses the role of LSD1 in cancer drug resistance and explores the potential of using LSD1 inhibitors in combination with other agents to overcome resistance.

Asciminib: a new therapeutic option in chronic-phase CML with treatment failure

Discovery of fragments inducing conformational effects in dynamic proteins using a second-harmonic generation biosensor

Summary: This study utilized a high-throughput SHG biosensor to detect conformational changes in proteins in real time and identify dynamic regions, successfully identifying conformational modulators of AChBP in a 1056 fragment library with hit rates ranging from 9-22% depending on the AChBP variant. The study also confirmed the reversible 1:1 interaction of fragments with AChBP and their binding at a previously described dynamic site, demonstrating the sensitivity of SHG in detecting fragments that induce conformational changes in proteins.
