Original Optimising cell-based bioassays via integrated design of experiments (ixDoE)- A practical guide

J. Solzin et al.

Summary: Despite being a more powerful approach than One Factor at a Time method, Design of Experiments (DoE) is not widely used in cell-based bioassay development. To address the reluctance of pharmaceutical manufacturers, we propose an integrated DoE (ixDoE) approach that optimizes resources and time by incorporating statistical inference from a single experimental set instead of the traditional multiple setups. This practical guide advocates for the innovative ixDoE approach and suitable statistical analysis strategy in bioassay development, spanning from basic research to biopharmaceutical industry.

CardioMotion: identification of functional and structural cardiotoxic liabilities in small molecules through brightfield kinetic imaging

William Stebbeds et al.

Summary: Cardiovascular toxicity is a major cause of drug failures and withdrawals in drug development. Current methods for early assessment of cardiovascular liabilities only capture acute functional cardiotoxicity but fail to detect structural cardiotoxicity that occurs after chronic exposure. CardioMotion is a novel label-free assay that records and assesses the beating of cardiomyocytes and can identify compounds that impact beating, providing a promising tool for early safety assessment.

Generation and maturation of human iPSC-derived 3D organotypic cardiac microtissues in long-term culture

Ece Ergir et al.

Summary: The study proposes a scaffold-free method to generate physiologically relevant human cardiac tissue models in vitro. These models exhibit self-organization, long-term culture ability, and contain multiple cell types and functional cardiac properties, making them a powerful platform for translational research in cardiovascular biology.

Sample size, power and effect size revisited: simplified and practical approaches in pre-clinical, clinical and laboratory studies

Ceyhan Ceran Serdar et al.

Summary: Calculating sample size in scientific studies is crucial for scientific contribution, impacting hypothesis and study design. Incorrect sample size may lead to inadequate results, time and cost losses, and ethical issues. This review aims to highlight the importance of sample size and provide researchers with tools to estimate it accurately.
