A Step Forward in the Characterization of Primary Brown Trout Hepatocytic Spheroids as Experimental Models

Transcriptomic and physiological analysis of endocrine disrupting chemicals Impacts on 3D Zebrafish liver cell culture system

Summary: In recent years, there have been efforts to develop in vitro platforms that mimic fish livers in order to better understand the effects of toxicants on aquatic vertebrates. Fish liver cell lines have emerged as a promising culture system in these platforms, as they complement existing in vitro tools and adhere to the 3Rs principles of animal testing. However, monolayer cell lines have shown lower transcriptional and physiological responses to toxic chemicals compared to freshly isolated primary cells. To overcome this challenge, a three-dimensional (3D) spheroid-based in vitro platform was utilized, which showed enhanced regulation of liver cells compared to monolayer cells. This platform was examined using the zebrafish liver (ZFL) cell line as a model system, and the 3D ZFL spheroids demonstrated increased synthesis of vitellogenin (Vtg) when treated with estrogenic chemicals. Transcriptome sequencing analysis confirmed that the 3D ZFL spheroids had greater transcriptional regulation of genes related to reproductive toxicological response and liver functions compared to monolayer cells. These findings contribute to the development of novel 3D in vitro platforms for screening harmful chemicals and improving understanding of liver toxicity mechanisms.

In vitro long term differentiation and functionality of three-dimensional bioprinted primary human hepatocytes: application for in vivo engraftment

Summary: In recent decades, the development of 3D in vitro cultures of primary human hepatocytes (PHHs) has become increasingly important for mimicking liver functions. This study utilized 3D bioprinting technology to print PHH in a GelMa matrix, resulting in the rapid organization of polarized hollow spheroids that remained viable for at least 28 days. The bioprinted PHH exhibited high differentiation and maintained liver-specific functions over time, including the activities of phase I and II biotransformation enzymes. The study also demonstrated the ability of the bioprinted structures to vascularize when implanted in mice. This model shows promise for generating human liver tissue and advancing biotechnological developments.

Fish hepatocyte spheroids - A powerful (though underexplored) alternative in vitro model to study hepatotoxicity

Summary: In vitro fish cell cultures are alternative models for in vivo toxicological studies. Two-dimensional cultures have limitations in longevity and maintaining in vivo cellular functions, and three-dimensional systems are proposed to better mimic in vivo effects. Fish hepatocyte spheroids, derived from liver cell lines or primary cultures, are the most used 3D model in fish. They closely resemble in vivo counterparts and have higher metabolic capacity compared to monolayer cultures.
