How the Innate Immune DNA Sensing cGAS-STING Pathway Is Involved in Apoptosis

Wanglong Zheng et al.

Summary: The cGAS-STING signaling axis is activated by cytosolic DNA, both non-self and self DNA. This pathway is used by the innate immune system to monitor invading pathogens and/or damage. It not only facilitates inflammatory responses and IFN production, but also activates other cellular processes like apoptosis.

Dual inhibition of innate immunity and apoptosis by human cytomegalovirus protein UL37x1 enables efficient virus replication

Yujie Ren et al.

Summary: The UL37 exon-1 protein encoded by human cytomegalovirus not only inhibits apoptosis but also suppresses the cGAS-STING immune pathway. It binds to TBK1 to disrupt the TBK1-STING-IRF3 interaction. The immunosuppressive and anti-apoptotic functions of UL37x1 are crucial for efficient HCMV infection.

The Porcine Cyclic GMP-AMP Synthase-STING Pathway Exerts an Unusual Antiviral Function Independent of Interferon and Autophagy

Sen Jiang et al.

Summary: The cGAS-STING pathway exerts antiviral activity through both IFN-dependent and IFN-independent mechanisms. This study reveals that the IFN-independent antiviral function of STING is independent of autophagy. In addition, cell apoptosis is associated with STING-mediated antiviral activity. The involvement of TBK1 and IRF3 in other STING activities, such as apoptosis, is also demonstrated.

The Porcine and Chicken Innate DNA Sensing cGAS-STING-IRF Signaling Axes Exhibit Differential Species Specificity

Sen Jiang et al.

Summary: This study reveals the species-specific differences in the cGAS-STING pathway in pigs and chickens, and identifies the motifs responsible for the species disparity. The study also demonstrates the specific and stringent role of pIRF3 in inducing IFN, which contributes to the inability of chSTING to induce IFN in mammalian cells.

How the Innate Immune DNA Sensing cGAS-STING Pathway Is Involved in Autophagy

Wanglong Zheng et al.

Summary: The cGAS-STING pathway is involved in triggering both canonical and non-canonical autophagy, which plays crucial roles in balancing innate immune responses, maintaining intracellular environmental homeostasis, alleviating liver injury, and limiting tumor growth and transformation.

TBK1 recruitment to STING activates both IRF3 and NF-κB that mediate immune defense against tumors and viral infections

Seoyun Yum et al.

Summary: This study shows that STING can function independently of type I interferons and autophagy, and that TBK1 recruitment to STING is essential for antiviral and antitumor immunity.

The crosstalk between the caspase family and the cGAS-STING signaling pathway

Yongai Xiong et al.

Summary: The manuscript discusses the role of cytosolic nucleic acid sensors during microbial infections and cell death, as well as the interaction between the caspase family and the cGAS-STING signaling pathway. This sheds light on novel therapeutic approaches targeting antiviral innate immunity, cancer, inflammatory diseases, and autoimmunity.
