Development of multifunctional drug delivery system via hot-melt extrusion paired with fused deposition modeling 3D printing techniques

Summary: The model of core-shell structured tablets is gaining interest in controlled-release and combinational drug delivery. This study used hot-melt extrusion and fused deposition modeling 3D printing techniques to develop a multifunctional core-shell model for controlled drug delivery. Different dosage forms could be successfully loaded into the produced shells. The designed model showed modified-release character and could improve formulation development for patient-centered medication.

Production of Bi-Compartmental Tablets by FDM 3D Printing for the Withdrawal of Diazepam

Summary: The present study aimed to prepare tablets with gradual doses of DZP using the dual-nozzle system of FDM 3D printers for the treatment of anxiety or acute alcohol withdrawal. Placebo and DZP-loaded filaments were prepared by hot-melt extrusion and used for printing bi-compartmental tablets. The printed tablets had good adhesion between layers and achieved the desired drug contents within the acceptance criteria.

Fabrication of timed-release indomethacin core-shell tablets for chronotherapeutic drug delivery using dual nozzle fused deposition modeling (FDM) 3D printing

Summary: This study developed timed-release indomethacin tablets for the treatment of early morning stiffness in rheumatoid arthritis, using two-nozzle fused deposition modeling (FDM) 3D printing. The tablets consisted of a drug-containing core and release-regulating shell, with different shell thicknesses. The tablets showed a delay of 4-8 hours depending on the shell thicknesses and released most of the drugs in 3 hours. The study explored the suitability of using FDM 3D printing with Bowden extrusion for producing personalized chronotherapeutic core-shell tablets.

Hot-melt extrudability of amorphous solid dispersions of flubendazole-copovidone: An exploratory study of the effect of drug loading and the balance of adjuvants on extrudability and dissolution

Summary: The FDA-approved anthelmintic, flubendazole, has potential to be used in the treatment of cancer and dry macular degeneration. However, its poor water solubility limits its use. In this study, the researchers evaluated the influence of adjuvants and drug loading on the development of an amorphous solid dispersion of flubendazole-copovidone using hot-melt extrusion. The results showed that hot-melt extrusion can be a viable option to obtain stable amorphous dispersions of flubendazole:copovidone, and the adjuvants had a notable influence on the dissolution rate.

QbD guided development of immediate release FDM-3D printed tablets with customizable API doses

Summary: The aim of this study was to develop a flexible 3D printed immediate release tablet using fused deposition modeling (FDM), which allows for adjusting the dose of the active pharmaceutical ingredient (API) by scaling the size of the dosage form. Polyvinyl alcohol-based filaments with elevated API content were prepared and their printability was evaluated. Quality by Design (QbD) approach and Design of Experiments (DoE) were employed for the tablet fabrication step. The effects of tablet design, size, and layer height settings on drug release and API content were investigated. The honeycomb configuration was found to be suitable for preparing customizable API doses in immediate release dosage forms. A predictive model was developed to optimize the printing variables.
