Summary: AS01 and ALFQ are two vaccine adjuvants composed of liposomes containing a mixture of natural substances. AS01 has been successfully used in malaria and shingles vaccines, while ALFQ is a similar adjuvant. We compared their chemical structures and physical characteristics and hypothesized that some side effects observed after liposome-based vaccine injections might be caused by the release of free fatty acids and lysophospholipids.

A Liquid Chromatography High-Resolution Tandem Mass Spectrometry Method to Quantify QS-21 Adjuvant and Its Degradation Products in Liposomal Drug Formulations

Summary: Identification and quantification of an active adjuvant and its degradation product are crucial for ensuring the safety and efficacy of drug formulations. A new LC-MS/MS method was developed to accurately quantify the active adjuvant QS-21 and its degradation product QS-21 HP in liposomal drug formulations. The method showed good specificity, sensitivity in the nanomolar range, and precise detection and quantitation for QS-21 and its degradation product.

Advances in giant unilamellar vesicle preparation techniques and applications

Additionally, the key applications of GUVs in studying membrane interface processes and constructing synthetic cells are described, along with a critical analysis of the pros and cons of different preparation technologies.

From LUVs to GUVs- How to Cover Micrometer-Sized Pores with Membranes

Summary: Pore-spanning membranes (PSMs) are a versatile tool for investigating membrane-confined processes. Fluorescence microscopy is most suitable for visualizing PSMs with micrometer-sized pores. However, the current method for preparing PSMs relies on the spreading of giant unilamellar vesicles (GUVs), which has limitations. In this study, a new method using droplet-stabilized GUVs generated in a microfluidic device was developed to produce PSMs from large unilamellar vesicles. The diffusion and mechanical properties of the resulting PSMs were analyzed and compared to PSMs derived from electroformed GUVs, showing that the GUV formation procedure has little impact on lipid diffusion and mechanical properties.
