Chitosan-based nanoscale systems for doxorubicin delivery: Exploring biomedical application in cancer therapy

Milad Ashrafizadeh et al.

Summary: Green chemistry has shown great promise in biomedical applications, particularly in cancer therapy. Chitosan-based nanoparticles (CS-NPs) serve as a promising carrier for anti-cancer drugs, overcoming drug resistance and enabling synergistic therapy. Various types of nanostructures can be modified with chitosan for efficient delivery of chemotherapeutic agents such as doxorubicin (DOX). The functionalization of CS-NPs with ligands and their pH-sensitive and redox-responsive characteristics further enhance their selectivity and drug release. Considering their high encapsulation efficiency and biocompatibility, significant progress is expected in the clinical translation of CS-NPs.

In situ forming alginate/gelatin hybrid hydrogels containing doxorubicin loaded chitosan/AuNPs nanogels for the local therapy of breast cancer

Anahita Alioghli Ziaei et al.

Summary: pH-sensitive in situ gelling hydrogels based on oxidized alginate and gelatin-containing doxorubicin (DOX) loaded chitosan/gold nanoparticles (CS/AuNPs) nanogels were synthesized. The CS/AuNPs nanogels had a size distribution of about 209 nm with a zeta potential of +19.2 mV and an encapsulation efficiency of around 72.6% for DOX. Rheological and texture analysis demonstrated that hydrogels containing β-GP and CS/AuNPs had higher mechanical properties. The release profile of DOX showed 99% and 73% release at pH = 5.8 and pH = 7.4, respectively. MTT cytotoxicity study confirmed the cytocompatibility of the prepared hydrogels on MCF-7 cells. Live/Dead assay revealed high cell viability on DOX-free hydrogels in the presence of CS/AuNPs nanogels, indicating the potential of the developed hydrogels for localized breast cancer treatment.

Structure-Property-Function Relationships of Iron Oxide Multicore Nanoflowers in Magnetic Hyperthermia and Photothermia

Enzo Bertuit et al.

Summary: Magnetite and maghemite multicore nanoflowers synthesized using modified polyol-mediated routes are highly effective nanoheaters in magnetic hyperthermia (MHT) and have also shown promising performances in photothermal therapy (PT) in the NIR-II biological window. The study investigates the influence of synthesis parameters on the size, shape, composition, cores, and crystallinity of the nanoflowers, establishing correlations between their structure, properties, and function in MHT and PT. Iron(III) and heating ramps are found to play a critical role in the formation of well-defined nanoflowers with multiple cores, with optical responses and photothermal properties depending on volume and structural disorder rather than stoichiometry. Defect concentration in the nanostructures affects optical behavior, with a transition in photothermal efficiency observed at a critical Urbach energy value.

Behaviour of a magnetic nanogel in a shear flow

Ivan S. Novikau et al.

Summary: This study investigates the shape, magnetic structure, and motion characteristics of magnetic nanogels under different shear rates using molecular dynamics simulations. The results show that, in a shear flow, the center of mass of the nanogels tends to be in the center of the channel and maintains a fixed distance from both walls. The monomers of the nanogel exhibit translation, rotation around the center of mass, and oscillation. This results in synchronized tumbling and wobbling of the nanogels accompanied by volume oscillations.
