Use of low-field NMR and rheology to evaluate the microstructure and stability of a poly(D,L-lactide-co-glycolide)-based W/O emulsion to be processed by spray drying

Ana Juric Simcic et al.

Summary: Drug-loaded emulsions for spray drying need to be optimized for their rheological behavior and stability. The study investigated the structure and stability of a water-in-oil emulsion containing vancomycin hydrochloride as the active ingredient. The addition of chitosan improved the long-term stability of the emulsion, and rheological data provided information about the velocity and shear rate/stress profiles during the spray drying process.

Controlling the Evolution of Selective Vancomycin Resistance through Successful Ophthalmic Eye-Drop Preparation of Vancomycin-Loaded Nanoliposomes Using the Active-Loading Method

El Tahra M. Ahmed et al.

Summary: Vancomycin is the foremost treatment choice for severe methicillin-resistant Staphylococcus aureus (MRSA) infections, but its therapeutic practice is limited by the emergence of vancomycin resistance. To overcome this, nanovesicles were used as a drug delivery system to enhance vancomycin therapy. The study successfully loaded vancomycin into liposomes using the ammonium sulfate gradient method, resulting in increased bactericidal effects against MRSA and heteroresistant vancomycin-intermediate S.aureous (h-VISA). Moreover, the loaded vancomycin prevented the development of resistance in MRSA. This finding suggests that vancomycin-loaded nanoliposomes can improve therapeutic use and control the emergence of vancomycin resistance.

Rheological and Viscoelastic Properties of Chitosan Solutions Prepared with Different Chitosan or Acetic Acid Concentrations

Paulo Jose do Amaral Sobral et al.

Summary: This study investigates the effect of chitosan and acetic acid concentrations on the rheological and viscoelastic properties of chitosan solutions. Chitosan concentration strongly affects the rheological properties, while the effect of acetic acid is less evident. All solutions behave as pseudoplastic fluids and exhibit viscoelastic properties.

Ultrasonic Atomization: New Spray Characterization Approaches

Miguel Panao

Summary: In particle engineering, spray drying is an important technique, and ultrasonic sprays have been found to be the best option for producing sprays with equal-sized droplets. This study explores new approaches to spray characterization and reveals the lack of universality in the relation between spray characteristics and Faraday waves. The results also show that drop size is influenced by liquid properties and flow rate, and that drop size diversity is a better indicator of ultrasonic spray characteristics. Additionally, lower flow rates lead to higher drop size diversity.

Formulation composition, manufacturing process, and characterization of poly(lactide-co-glycolide) microparticles

Kinam Park et al.

Summary: Injectable long-acting formulations, particularly those based on poly(lactide-co-glycolide) (PLGA), have been utilized for systemic drug delivery for up to 6 months, but the development of clinical products and generics has been slow. The limited understanding of PLGA polymers and the complex variables involved in composition and manufacturing processes hinder the control over formulation properties and drug efficacy. Factors such as formulation composition, manufacturing processes, and surface morphology play crucial roles in determining the pharmacokinetic profile of the formulations, highlighting the importance of proper characterization and control of these parameters for desired drug release kinetics and efficacy.
