An Update on the Use of Allometric and Other Scaling Methods to Scale Drug Clearance in Children: Towards Decision Tables

Increasing application of pediatric physiologically based pharmacokinetic models across academic and industry organizations

Summary: There has been a significant increase in the use of physiologically based pharmacokinetic (PBPK) models in the field of pediatrics during the past 20 years. This study provides a detailed overview of the growth and various applications of pediatric PBPK models. The results show that the use of P-PBPK modeling has grown rapidly, primarily in clinical settings and drug development. The study also highlights the secondary applications of dose selection, pharmacokinetic prediction, and model parameter identification. However, the study suggests that there is still potential for further application of P-PBPK models in other areas such as pediatric formulations assessment, toxicology, and trial design.

A meta-analysis of protein binding of flucloxacillin in healthy volunteers and hospitalized patients

Summary: The study aimed to develop a mechanistic protein-binding model for predicting the unbound flucloxacillin concentrations in different patient populations. The model outperformed the calculation with a fixed unbound fraction of 5% in terms of accuracy and prediction error. A permeability- and perfusion-based PBPK model for improved prediction of concentration-time profiles

Summary: PermQ, a new physiologically based pharmacokinetic modeling framework, has been developed to improve predictions of concentration-time profiles of drugs. The model includes permeability into and out of capillaries, cell membranes, and intracellular lipids. Results show that the PermQ model reduces errors in V-ss and t(1/2) by 37% and 43% respectively, and improves C-t profiles by 43%, especially for basic drugs.

Clinical Pharmacokinetics and Pharmacodynamics of Cefepime

Summary: Cefepime is a broad-spectrum antibiotic with activity against various bacteria. It can be administered through infusion or prolonged infusion. In healthy adults with normal renal function, cefepime is widely distributed and about 85% of the dose is excreted unchanged in the urine, with an elimination half-life of 2-2.3 hours. The pharmacokinetics of cefepime can be altered under certain pathophysiological conditions, resulting in high inter-individual variability and posing challenges for population dosing approaches. Therapeutic drug monitoring of cefepime may be beneficial in certain special patients.

An Overview of the Protein Binding of Cephalosporins in Human Body Fluids: A Systematic Review

Summary: This systematic literature review examines the protein binding of cephalosporins in human body fluids, finding variations in protein binding among different fluids and patient categories. Optimizing Meropenem Therapy for Severe Nosocomial Infections in Neonates
