Summary: Iron is an essential cofactor in the biosynthesis of cellular components, and the ergosterol biosynthetic pathway in yeast is dependent on iron. The expression of ERG genes is tightly controlled by iron availability, which regulates sterol and heme levels. Transcription factors Upc2 and Ecm22 activate ERG gene expression during iron deficiency, while Hap1 triggers transcriptional repression mediated by Tup1. The combined regulation of these factors allows for fine-tuning of ERG transcript levels during iron deficiency.

Synergistic Antifungal Effect of a Combination of Iron Deficiency and Calcium Supplementation

Summary: Millions of immunocompromised people are at a higher risk of developing severe fungal diseases. The limited number of antifungals and the emergence of antimicrobial resistance call for new strategies against invasive fungal infections. This study demonstrates that calcium supplementation combined with iron deficiency inhibits the growth of human fungal pathogens, including Aspergillus fumigatus, Candida albicans, and Cryptococcus neoformans. Iron deficiency induces a nonspecific calcium uptake response, leading to toxic levels of metal. The findings suggest that a microenvironment with excess calcium and limited iron is an efficient strategy to curb the growth of fungal pathogens, especially for drug-resistant isolates.

The Cryptococcus neoformans Flc1 Homologue Controls Calcium Homeostasis and Confers Fungal Pathogenicity in the Infected Hosts

Summary: This study reveals the importance of the Flc1 protein in maintaining calcium homeostasis and vacuolar biogenesis in Cryptococcus neoformans. Deletion of Flc1 leads to an increase in cytosolic calcium concentration, an elevation of Crz1 transcription factor in the nucleus, and an aberrant accumulation of cell wall chitin. Flc1 is also involved in vacuolar fusion and capsule formation. The absence of Flc1 results in the loss of survival ability and virulence in Cryptococcus.
