UniProt: the Universal Protein Knowledgebase in 2023

Summary: The UniProt Knowledgebase aims to provide comprehensive, high-quality, and freely accessible protein sequences annotated with functional information. The database has expanded its data processing pipeline and website to accommodate the increasing information content, with over 227 million sequences and plans to include a reference proteome for each taxonomic group. Detailed annotations are extracted from the literature to update or create reviewed entries, while unreviewed entries are supplemented with annotations from automated systems. The new website, https://www.uniprot.org/, offers enhanced user experience and easy access to data, including AlphaFold structures and improved protein subcellular localization visualizations.

Systematic pan-cancer analysis identifies transmembrane protein 158 as a potential therapeutic, prognostic and immunological biomarker

Summary: The purpose of this study was to investigate the expression significance, predictive value, immunologic function, and biological role of transmembrane protein 158 (TMEM158) in the development of pan-cancer. Our findings revealed that TMEM158 was highly expressed in various cancer tissues and was associated with prognosis. Moreover, TMEM158 was significantly correlated with tumor mutational burden (TMB), microsatellite instability (MSI), and tumor immune cell infiltration in multiple cancers. TMEM158 may serve as a significant predictor of cancer prognosis and modulate immune responses to various types of cancer.

TMEM45B Interacts with Sindbis Virus Nsp1 and Nsp4 and Inhibits Viral Replication

Summary: This study found that the ISG product TMEM45B inhibits the replication of Sindbis virus (SINV). TMEM45B interacts with the viral nonstructural proteins Nsp1 and Nsp4, inhibiting the translation and promoting the degradation of SINV RNA. In addition, TMEM45B also interacts with Nsp1 and Nsp4 of chikungunya virus (CHIKV), suggesting its potential role in inhibiting the replication of other alphaviruses.

Summary: Preliminary results suggest that TMEM180 may promote colon cancer growth, without significant effects on oxygen consumption or phosphorylated protein expression. However, glycolysis differs significantly between SW480 cells and TMEM180-knockdown cells. NGS analysis indicates that TMEM180 promotes enzyme expression in the nitric oxide synthesis system, potentially contributing to glucose and glutamine metabolism and cancer growth. Further basic studies of the TMEM180 molecule are warranted based on these findings.
