Although supplementation of vitamin D is considered safe and inexpensive, the effects on various diseases are not well-established. Future studies need to carefully consider the challenges in assessing the potential benefits of vitamin D, such as the evaluation of the whole vitamin D endocrine system and use of adequate dosing.

Vitamin D insufficiency and serum levels related to the incidence and stage of cutaneous melanoma: a systematic review and meta-analysis

Summary: This study evaluated the relationship between vitamin D insufficiency and 25-hydroxy vitamin D levels with the incidence and stage of Cutaneous melanoma (CM). The results showed a significant association between vitamin D level <20 ng/dl and incidence of CM; lower mean vitamin D level and Breslow depth >1 mm were also significantly related; and vitamin D level >20 ng/dl was associated with a lower Breslow depth <1 mm.

Vitamin D: 100 years of discoveries, yet controversy continues

Summary: In the past 100 years, significant progress has been made in vitamin D research, including the cure of rickets, discovery of vitamin D compounds, advances in vitamin D molecular biology, and understanding endocrine control of vitamin D metabolism. Clinical trials have been conducted to explore the effects of vitamin D in preventing various diseases, but did not meet expectations. Some trials also revealed unexpected adverse events, such as increased fractures and falls in older people. More research is needed to determine the safety and efficacy of high-dose vitamin D supplementation.

Enzymatic activation in vitamin D signaling-Past, present and future

Summary: Vitamin D signaling plays a crucial role in maintaining calcium balance for bone health and has various functions in different tissues. Disturbances in vitamin D signaling are associated with numerous diseases. This review focuses on the identification of bioactivating enzymes and their genes involved in the production of active vitamin D metabolites. It evaluates species- and tissue-specific expression, enzyme kinetics, substrate specificity, and the consequences of gene mutations. The roles of different vitamin D receptors and alternative bioactivation pathways are also discussed. Although significant progress has been made in understanding vitamin D bioactivation enzymes, further research is needed to fully comprehend the pleiotropic effects and the mechanisms of enzymatic activation in vitamin D-induced responses.

However, overall survival rates have improved relatively modestly. Melanoma exhibits heterogeneity and transcriptional plasticity, enabling it to adapt and escape even advanced treatments. Although our understanding of melanoma biology and genetics has advanced, debates continue regarding the cell of origin for melanoma. Animal models and single-cell sequencing approaches offer new avenues of research in addressing this question.
