Bcr-TMP, a Novel Nanomolar-Active Compound That Exhibits Both MYB- and Microtubule-Inhibitory Activity

Maria V. Yusenko et al.

Summary: This study introduces a novel MYB inhibitory compound called Bcr-TMP, which shows potential therapeutic effects on leukemias and cancers dependent on deregulated MYB activity. Bcr-TMP down-regulates MYB target genes, induces cell death and differentiation, and exhibits stronger anti-proliferative effects on MYB-addicted primary cells. Additionally, Bcr-TMP acts through p300 and has anti-microtubule activity. Further research is needed to understand its mechanism of action and therapeutic potential.

Report Single-cell RNA sequencing identifies a paracrine interaction that may drive oncogenic notch signaling in human adenoid cystic carcinoma

Anuraag S. Parikh et al.

Summary: This study utilized single-cell RNA sequencing to analyze salivary adenoid cystic carcinoma (ACC) at the cellular level, revealing expression heterogeneity in myoepithelial and luminal cells and differential expression of Notch signaling-related genes in myoepithelial cells. These findings may provide an explanation for the biphasic nature of low-and intermediate-grade ACC and offer insights for new therapeutic strategies.

Current opinions on diagnosis and treatment of adenoid cystic carcinoma

Yan Fang et al.

Summary: Adenoid cystic carcinoma (ACC) is a rare malignant tumor derived mainly from the salivary glands that displays contradictory characteristics of slow growth but aggressive progression. ACC of the head and neck (ACCHN) has a propensity for perineural invasion and is challenging to treat. This review aims to provide updated insights into ACCHN and discuss imaging diagnosis, pathological features, and therapeutic strategies.

A Randomized Phase 2 Study of Pembrolizumab With or Without Radiation in Patients With Recurrent or Metastatic Adenoid Cystic Carcinoma

Umair Mahmood et al.

Summary: This study evaluated the safety and efficacy of pembrolizumab with or without radiation therapy in patients with adenoid cystic carcinoma. While no objective responses were observed, a majority of patients achieved stable disease with decreased tumor growth rate and half experienced clinical benefit exceeding 6 months. Favorable local responses were also observed within the radiation field, highlighting the need for additional strategies to delay progression and improve response.

J-f He et al.

Summary: This study retrospectively evaluated the clinicopathological aspects and prognostic factors of 40 patients with adenoid cystic carcinoma of the intraoral minor salivary glands. Results showed that the tumor mainly occurred in the palate, with common findings of positive surgical margins and perineural invasion. Adjuvant postoperative radiotherapy played a positive role in improving patient survival, while tumor size and positive surgical margins were important factors in predicting local recurrence.

Proteasome inhibitors suppress MYB oncogenic activity in a p300-dependent manner

Maria V. Yusenko et al.

Summary: Studies on MYB in human malignancies have identified it as a potential drug target for AML and ACC. While transcription factors are often considered hard to target, recent developments have shown successful targeting of MYB with low molecular weight compounds. The use of proteasome inhibitors to suppress oncogenic MYB activity opens up new possibilities for therapeutic approaches in MYB-driven cancers.

MYB oncoproteins: emerging players and potential therapeutic targets in human cancer

Ylenia Ciciro et al.

Summary: MYB transcription factors are highly conserved from plants to vertebrates, and their aberrant activation in humans may lead to cancer. Recent detection of MYB gene rearrangements and mutations in human cancer provides smoking gun evidence of the involvement of MYB family members in human malignancies. Studies linking the activity of MYB family members to human cancers and experimental therapeutic interventions tailored for MYB-expressing cancers are highlighted in this review.
