Summary: The study aimed to investigate the clinical relevance of reactive oxygen species (ROS) in the breast cancer tumor microenvironment. The results showed that high ROS levels were significantly associated with cell proliferation-related genes, pro-cancer genes, immune-related genes, infiltrated immune cells, and increased cytolytic activity. High ROS was also associated with intratumor heterogeneity, mutation rates, immune response, and worse survival outcomes.

It also regulates the niche of cancer stem cells. Translating this combination therapy to bedside applications is feasible.

Hyperbaric oxygen and radiation therapy: a review

Summary: Hyperbaric oxygen therapy (HBOT) has been used for decades as a treatment for radiation injuries, with evidence showing that at pressures exceeding 2 absolute atmospheres (ata), HBOT can reduce late-onset radiation injuries to various areas. HBOT also appears to prevent osteoradionecrosis and enhance the effectiveness of radiation in certain types of tumors. Promising results have been achieved in the local control of high-grade gliomas with HBOT at 2 ata.

The relationship between expression of PD-L1 and HIF-1α in glioma cells under hypoxia

Summary: The study showed a positive correlation between PD-L1 and HIF-1α in glioma, with their overexpression associated with poor survival. HIF-1α was found to induce PD-L1 expression under hypoxic conditions, which could be abrogated by inhibiting HIF-1α. Combination therapy of HIF-1α inhibitor and anti-PD-L1 antibody demonstrated enhanced tumor growth suppression and improved immune activation.

HBO enhanced PD-1 Ab delivery and T cell infiltration, disrupting immunosuppression and promoting robust immune responses to inhibit tumor relapses. Such enhanced efficacy was observed in solid tumors in rodents and hepatocellular carcinoma patients, highlighting the potential of HBO-PD-1 Ab combination therapy in stroma-rich solid tumors.
