Summary: The study demonstrates a highly efficient single-animal encapsulation unit for controlling the chemical environment in droplets, as well as an automation scheme for executing complex protocols. By careful use of interfacial forces, the system enables the generation of chemical gradients inaccessible in previous multiphase systems. This platform allows for large-scale screening of complex dynamic phenotypes in small animals.

Splitting of droplet with different sizes inside a symmetric T-junction microchannel using an electric field

Lubrication effects on droplet manipulation by electrowetting-on-dielectric (EWOD)

Summary: In this study, droplet-migration characteristics on oil-infused electrowetting-on-dielectric substrates were investigated. It was found that the droplet velocity is scaled as U approximate to E-2, which differs from a relationship of U approximate to E-3 predicted from the dominant drag force. The achieved high velocity is explained by a lubrication-flow effect.

A droplet-based microfluidic platform enables high-throughput combinatorial optimization of cyanobacterial cultivation

Summary: A droplet-based microfluidic platform was used to systematically optimize cultivation parameters for cyanobacteria. Three different unicellular cyanobacteria were successfully grown, and one-dimensional and two-dimensional screenings were performed to determine key nutrient requirements. The results suggested that nitrate and/or phosphate may be limiting nutrients in standard cultivation media. Improved medium composition led to a 15.7% increase in biomass yield.

While flow cytometry relies on cell surface and intracellular biomarkers, it lacks spatial and temporal phenotypes and activities of cells. On the other hand, droplet microfluidics integrates flow cytometry to link cell phenotypes to genotypes at the single-cell level, offering a powerful tool for cellular assays.
