Toward a new generation of vaginal pessaries via 3D-printing: Concomitant mechanical support and drug delivery

Summary: This study explores the use of 3D printing to customize the mechanics and drug release of vaginal pessaries made from pharmaceutical grade thermoplastic polyurethanes (TPU). Soft hydrophobic TPU was found to be the most promising candidate for pessary customization, providing a wide range of mechanical parameters and allowing self-insertion. Hydrophobic TPUs also showed superior drug release performance, closer to the target value. 3D printing of TPU-based pessaries is an innovative strategy to create customized products that offer both mechanical support and pharmacological therapy.

Tailoring amlodipine release from 3D printed tablets: Influence of infill patterns and wall thickness

Summary: This study investigated the impact of infill patterns and wall thickness on drug release of 3D-printed tablets, and the potential of using excipients to tailor drug release. Results showed that zigzag infill pattern and SSG excipient led to faster drug release, while HPMC HME 4 M had minimal effect on release rate. Additionally, decreasing wall thickness was associated with higher drug release. Proper selection of excipients and adjustment of infill patterns and wall thickness are important for tailoring drug release in FDM 3D printing.
