Improved Method for Dental Pulp Stem Cell Preservation and Its Underlying Cell Biological Mechanism

Genome-Wide Screening of Differentially Expressed Genes and their Potential Associations with Aging Dental Pulp Stem Cells

Summary: In this study, the differences in expression of long non-coding RNAs (lncRNAs) and messenger RNAs (mRNAs) between aging and young dental pulp stem cells (DPSCs) were investigated using RNA sequencing, pathway enrichment, and gene ontology analysis. The results revealed 14 differentially expressed mRNAs and 7 differentially expressed lncRNAs between aging and young DPSCs. Furthermore, hsa-miR-6724-5p was identified as a key regulator in the aging process of DPSCs, with its target genes involved in the dopaminergic synapse. These findings suggest that the aging of DPSCs is associated with differentially expressed genes (DEGs) related to the dopaminergic synapse, but further investigation is needed to understand the specific functions and internal relationships of these DEGs.

Long-term hypoxia inhibits the passage-dependent stemness decrease and senescence increase of human dental pulp stem cells

Summary: This study verified the effects of long-term hypoxic culture on human dental pulp stem cells (hDPSCs). The results showed that hypoxic culture maintained the morphology, phenotype, and proliferation activity of hDPSCs, while reducing cell senescence. Hypoxic culture also increased the expression of stemness markers and decreased the expression of senescence-related genes. Additionally, hypoxic culture suppressed spontaneous and induced osteogenic and adipogenic differentiation. These findings highlight the beneficial effects of long-term hypoxic culture on hDPSCs' biological characteristics.
