Neuroimmune Interactions in Fetal Alcohol Spectrum Disorders: Potential Therapeutic Targets and Intervention Strategies

Molecular Markers in Maternal Blood Exosomes Allow Early Detection of Fetal Alcohol Spectrum Disorders

Summary: Prenatal alcohol exposure can lead to fetal alcohol spectrum disorders (FASD), which include developmental abnormalities in the eyes, face, and brain, as well as neurobehavioral deficits. However, these abnormalities cannot be detected early in pregnancy using current imaging techniques. This study found that the levels of certain markers, particularly myelin basic protein (MBP), in fetal brain-derived exosomes (FB-E) in maternal blood were highly correlated with morphological abnormalities associated with alcohol exposure. These findings suggest that FB-E markers, such as MBP, may be used as predictors of FASD.

Dose-related shifts in proteome and function of extracellular vesicles secreted by fetal neural stem cells following chronic alcohol exposure

Summary: Accumulating evidence suggests that extracellular vesicles (EVs) mediate endocrine functions and the pathological effects of neurodevelopmental perturbagens such as ethanol. This study identified overrepresented proteins and signaling pathways in EVs secreted by fetal murine cerebral cortical neural stem cells (NSCs) using mass-spectrometry, and found that the EV proteomes differ significantly from parental NSCs. Ethanol exposure resulted in selective protein sequestration in EVs and depletion in parental NSCs, as well as overrepresentation of certain pathways in EVs. Transfer of untreated EVs to naive cells had a growth-inhibitory effect, while EVs derived from ethanol-treated NSCs had a diminished effect. These findings suggest that NSCs secrete EVs with a distinct proteome that may play a role in inhibiting the growth of recipient cells, and that ethanol exposure affects the efficacy of these EVs.
