m6A modification: recent advances, anticancer targeted drug discovery and beyond

Li-Juan Deng et al.

Summary: Abnormal N6-methyladenosine (m6A) modification is closely related to cancer development, and finding targeted anticancer drugs is important. Traditional herbs and computer-synthesized compounds are reasonable sources for these drugs. Artificial intelligence has advantages in discovering m6A-targeting anticancer drugs.

Low expression of IGFBP4 and TAGLN accelerate the poor overall survival of osteosarcoma

Yue Xi et al.

Summary: By using the GEO database and bioinformatics analysis, we identified IGFBP4 and TAGLN as key genes in osteosarcoma. Osteosarcoma patients with high expression levels of IGFBP4 and TAGLN had better overall survival times. Correlation analysis showed significant associations between IGFBP4 and tumor metastasis, Enneking stage, and TAGLN. Experimental verification demonstrated down-regulation of IGFBP4 and TAGLN expression in osteosarcoma tissue. Support Vector Machine analysis revealed a strong relationship among the expression of IGFBP4, TAGLN, and osteosarcoma.

Novel insights into the interaction between N6-methyladenosine methylation and noncoding RNAs in musculoskeletal disorders

Juanjuan Han et al.

Summary: This review summarizes the molecular regulatory mechanism of noncoding RNA (ncRNA) N6-methyladenosine (m6A) modification in musculoskeletal disorders (MSD). Numerous studies have confirmed the mutual regulation between m6A and ncRNAs, and ncRNA m6A modification plays an essential role in the pathophysiological process of MSD.

Large-Scale Multi-omic Analysis of COVID-19 Severity

Katherine A. Overmyer et al.

Summary: The study conducted RNA-seq and high-resolution mass spectrometry on blood samples from COVID-19-positive and COVID-19-negative patients, identifying molecular features associated with disease severity and outcomes, offering pathophysiological insights and therapeutic suggestions. The observed dysregulation of platelet function, blood coagulation, acute phase response, and endotheliopathy further illuminated the unique COVID-19 phenotype. The findings are presented through a web-based tool enabling interactive exploration and machine learning prediction of COVID-19 severity.

Comparative evaluation of two methods for LC-MS/MS proteomic analysis of formalin fixed and paraffin embedded tissues

Katarina Davalieva et al.

Summary: This study compared two different sample preparation methods for shotgun proteomic analysis of FFPE tissues and found that RapiGest identified more proteins, exhibited higher technical reproducibility, and showed greater similarity to fresh tissues compared to FASP. Therefore, RapiGest is recommended for biomarker and quantification studies due to its better performance in protein identification and preservation in FFPE tissues.

Proteomic comparison between different tissue preservation methods for identification of promising biomarkers of urothelial bladder cancer

Alberto Valdes et al.

Summary: Samples preserved by different methods, FFPE and OCT, show differences in protein analysis results using MS technology, with preservation method being a main source of variation. Proteins related to mitochondrial function are overrepresented in OCT data but missing in FFPE data. The use of MS technology for biobank samples is supported and further evaluation of proteins as biomarkers is encouraged.

Regulatory Role of N6-methyladenosine (m6A) Modification in Osteosarcoma

Yujie Zhang et al.

Summary: Osteosarcoma is the most common primary bone malignancy, typically seen in childhood or adolescence. Clinical outcomes for patients with osteosarcoma are usually poor due to the aggressive nature of the disease and limited treatment advances in the past four decades. Dysregulation of m(6)A-related factors in multiple osteosarcoma processes has been observed, indicating potential for future research and clinical applications.

Summary: The study demonstrates that arginine methylation of hnRNPK suppresses apoptosis of U2OS cells by interfering with the interaction with RNA helicase DDX3, while the DDX3-hnRNPK interaction may serve as a target for promoting apoptosis in osteosarcoma cells.

An update on the roles of circular RNAs in osteosarcoma

Zheng Li et al.

Summary: Osteosarcoma, the most common primary bone malignancy, is believed to arise from bone-forming mesenchymal stem cells, with aberrant activation of oncogenes and inactivation of tumor suppressor genes playing a key role in its pathogenesis. In addition to protein-coding genes, non-coding RNAs, especially circRNAs, have been found to be crucial in the initiation and progression of osteosarcoma. CircRNAs, single-stranded RNAs that form covalently closed loops, function as important regulatory elements of the genome and have been shown to play critical roles in osteosarcoma progression.
